摘要
目的:筛选参与口腔扁平苔藓(oral lichen planus,OLP)上皮细胞凋亡以及炎症反应相关的胞内关键转录信号蛋白。方法:选取OLP患者8例与正常对照者6例,分别收集口腔损害黏膜组织与正常颊黏膜组织及外周血5 m L,并提取各组黏膜组织与外周血单个核细胞的蛋白及测定蛋白浓度。使用奥德赛~■红外成像系统扫描并利用LI-COR~■ Image Studio v2.0芯片分析软件测量各组平均RFU值后统计分析。结果:与正常对照和萎缩糜烂型OLP组比较,网纹型OLP损害组织中p38 MAPK(Thr180/Tyr204)明显升高(P<0.001,P<0.001)。而STAT3(Ser727)在正常对照组、网纹型以及萎缩糜烂型OLP组织中的表达水平依次升高(P<0.001,P=0.002)。此外,在外周血中,与正常对照组比较,OLP患者p38 MAPK(Thr180/Tyr204)与STAT3(Ser727)表达均明显升高。结论:STAT3(Ser727)与p38 MAPK(Thr180/Tyr204)可能是OLP疾病发生发展过程中与临床分型密切相关的关键转录信号蛋白。
Objective : To screen the key transcription signaling proteins, associated with the epithelium apoptosis and inflammation in the OLP, and to understand the pathogenesis of OLP. Methods : OLP patients ( n = 8 ) and the normal control group( n = 6 )were collected ,then oral mucosa lesion, normal buccal umcosa and peripheral blood from each group were captured. The concentration of the protein from each specimen was determined after it was extracted. Samples were added in the plate according to the kit instruction, then the results were scaned by using the Odyssey~ infrared imaging system. Finally, the average RFU value of every sample was analyzed by using the LI-COR~ Image Studio v2. 0 chip analysis software. Results: p38 MAPK( Thr180/Tyr182 )expression increased significantly in the reticular OLP( both P 〈 0. 001 )compared to the control and erosive mucosa. However, in the erosive lesions, compared with the control and reticular mucosa, STAT3 (Ser727)expression was significantly elevated (P 〈 0. 001 and P = 0. 002 respectively). Meanwhile, STAT3 (Ser727)expression in the reticular OLP was higher compared to the normal control( P = 0. 001 ). In addition, the expression of p38 MAPK and STAT3 in- creased significantly in the PBMCs from the OLP patients, compared with those from the normal controls (P = 0. 001 and P = 0. 006 respectively). Conclusion :p38 MAPK (Thr180/Tyr182)and STAT3 (Ser727), as important signal transduction factors, might be involved in the process of OLP, especially account for the different clinical manifestations of OLP.
