摘要 目的观察mTOR抑制剂雷帕霉素对肺癌LK2细胞生长的影响。方法 LK2细胞在0.25、0.5、1、2、4、8μmol/L不同浓度的雷帕霉素组以及0μmol/L雷帕霉素空白对照组作用下,采用MTT比色法检测细胞生长情况,流式细胞术检测细胞的凋亡改变。结果雷帕霉素能够抑制LK2细胞增殖,且具有时间-剂量效应关系。流式细胞仪检测提示,雷帕霉素各浓度组均可以促进肺癌细胞LK2凋亡,且随着雷帕霉素浓度的升高,LK2细胞凋亡率逐渐增加。结论雷帕霉素对肺癌LK2细胞有抑制作用。 Objective To observe the effect of rapamycin on the growth of LK2 cells in lung cancer. Methods LK2 cells were evaluated by MTT assay and flow cytometry to detect their growth and apoptosis in different concen- trations of rapamycin groups and blank control group. Results Rapamycin could inhibit the proliferation of LK2 cells and had a time-dose effect relationship. Flow cytometry result showed that the apoptosis ratio of LK2 cells increased with the increase of rapamycin concentration in the experiment. Conclusion Rapamycin has an inhibitory effect on lung cancer LK2 cells.
