摘要
建立了以高效液相色谱法测定盐酸沃尼妙林关键中间体二甲基半胱胺截短侧耳素含量的检测方法,色谱条件为SepaxC18色谱柱(150 mm×4.6 mm,3μm)流动相为乙腈-磷酸缓冲液(p H2.5)(V/V=40∶60),流速为1 m L/min,检测波长为205 nm。对该检测方法进行了系统适用性、线性、精密度、重现性稳定性、检测限、定量限以及回收率的考察,结果均良好,提示该方法可以用于盐酸沃尼妙林关键中间体的含量检测。
An HPLC method for the determination of dimethyl cysteaminyl pleuromulin, a key valnemulin hydrochloride, was established. The mobile phase consisted of acetonitrile-phosphate (V/V=40 : 60) with a flow rate of 1 mL/min and a detection wavelength of 205 nm. The intermediate of buffer ( pH2.5 ) chromatographic conditions were SepaxC18 (150 mmx4.6 mm, 3 μm). The linearity, precision, reproducibility, detection limit, quantitation limit and recovery rate of the method were tested. The results were reproducible and could be applied to the content control of the key intermediates of valnemulin hydrochloride.
