热带无爪螨主要变应原Blo t 21抗原表位的预测 被引量:2

摘要 目的预测热带无爪螨主要变应原Blo t 21的抗原表位。方法利用DNAStar Protean软件分析热带无爪螨主要变应原Blo t 21的二级结构和表面特性,如理化性质、亲水性、表面可及性、柔韧性等,预测Blo t 21蛋白的B抗原表位和T抗原表位,同时利用Bepi Pred在线分析预测其可能的B细胞抗原表位。结果 Blo t 21蛋白的二级结构以α螺旋为主,存在7个潜在的B抗原表位位点,可能的B抗原表位区域有:2-11,19-28,35-54,55-70,72-83,96-112,123-129氨基酸残基或其附近。含有8个潜在的T细胞抗原表位区域:13-17,22-31,37-46,51-64,69-75,80-84,95-99,102-105氨基酸残基或其附近。结论应用Protean软件和Bepi Pred在线预测了热带无爪螨主要变应原Blo t 21的抗原表位,为进一步研究Blo t 21变应原的结构与功能,研制针对热带螨的预防性和治疗性疫苗、开发相应的诊断试剂盒奠定了基础。 Objective To predict the epitopes of the major allergen Blo t 21 from Blomia tropicalis. Methods The secondary structure and surface properties of Blo t 21 allergen were analyzed by DNAStar Protean software ,such as physical and chemical characters,hydrophilicity,surface probability and flexibility. Then the B cell epitopes and T cell epitopes of Blo t 21 were predicted by DNAStar software. At the meanwhile ,the Bcell epitopes for Blo t 21 were predicted with the online software BepiPred. Results The secondary structure of Blo t 21 allergen was rich in alpha helix. It contained seven potential B cell epitopes located at 2-11,19-28,35-54,55-70,72-83,96-112,123-129 amino acid residues or nearby. There were eight potential T cell epitopes located at 13-17,22-31,37-46,51-64,69-75,80-84,95-99,102-105 amino acid residues or nearby. Conclusion The epitopes of the major allergen Blo t 21 from Blomia tropicalis were predicted by DNAStar Protean and BepiPred software. The results could provide a base for the studies of structure and function of Blo t 21 allergen,development of the preventive and therapeutic vaccines for Blomia tropicalis,and diagnostic kit for the detection of allergies induced by Blomia tropicalis.
