摘要
应用流式细胞术(FCM)和Feulgen-DNA法显示分裂指数,分析不同浓度山豆根对Eca-109细胞的作用。流式细胞光度术分析结果表明,实验组细胞DNA指数(DI=2)显著低于对照组(DI=2.5),两者相比差异均有极其显著性意义(P<0.01)。细胞DNA组方图G_0/G_1期细胞呈增高趋势,S期细胞均减少,G_2M期细胞均增高,与对照组比差异均有显著性(P<0.01,P<0.05)。Feulgen-DNA染色显示细胞分裂指数与小剂量药物作用时间呈负相关,大剂量药物作用下分裂指数明显下降。这说明山豆根对Eca-109细胞DNA合成有显著的抑制作用。
Objective To study on influences of different dosages of Sophora Tonkinensis Gagnep on the Eca - 109 cell line.Methods Division index was tested by flow cyctometry (FCM) and Feulgen-DNA.Results By using FCM, experimental cell' s DNA index (DI = 2) were lower than ID in the control group (DI = 2.5) .There was a significant difference between them (P< 0.01) .Cells on G0/G1 and G2M stage were higher than those in the control group, cells on s stage were lower than those in the control group in cell DNA histogram (P<0.05, P<0.01) .Cell division index negatively correlated with time of low - dose Sophora Tonkinensis Gagnep by Feulgen - DNA stain. Conclusion Sophora Tonkineensis Gagnep has a significant inhibition on the DNA synthesis of Eca - 109 cell.
