摘要
目的探讨深圳市人群中IGF2BP2(rs1470579)及PTPRD(rs17584499)的单核苷酸多态性位点与妊娠期糖尿病(gestational diabetes mellitus,GDM)易感性的相关性。方法本研究于2015年6月10日至2015年12月30日,选取深圳市宝安区妇幼保健院505名孕妇纳入研究,其中研究组153名,对照组352名。留取研究对象2 mL外周静脉血,运用Taqman探针法对505例研究对象标本进行2个SNP位点PCR扩增、基因型测定,分析候选SNP位点在研究组及对照组中的基因型和等位基因分布频率,运用SPSS18.0、SNPstats、MDR和Microsoft Excel 2010软件进行数据分析和图表制作。结果 1.研究组(n=153)孕妇平均年龄为(27.81±4.06)岁,对照组(n=352)孕妇平均年龄为(27.88±3.86)岁,两组间差异无统计学意义(t=0.33,P=0.74);研究组孕前BMI为(20.32±2.57)kg/m^2,对照组孕前BMI为(20.29±2.44)kg/m^2,两组间差异无统计学意义(t=0.45,P=0.33);2.目标基因位点SNPs基因型在对照组分布的频率均符合HW遗传平衡(P>0.05);3.在显性模型、隐性模型、共显性模型、超显性模型、加性模型中,均未发现IGF2BP2 rs1470579及PTPRD rs17584499多态性与GDM易感性存在关联。结论未发现所研究的2个SNPs与GDM易感性存在相关性。
Objective To explore the association between two single nucleotide polymorphisms( SNPs),including rs1470579 locus in IGF2BP2 gene and rs17584499 locus in PTPRD gene, and the genetic susceptibility of gestational diabetes mellitus( GDM) in Shenzhen city population. Methods During June 10 to December 30 in 2015,a total of 153 GDM and 352 normal glucose tolerant pregnant women in Maternal and Child Health Hospital,Bao'an,Shenzhen were enrolled. 2 ml fasting venous blood were collected,and TaqM an genotyping technology was used to determine the 7 SNPs genotypes. The relationships of the polymorphisms and GDM susceptibility were calculated. In addition, multifactor dimensionality reduction was applied to analyze the impact of gene-gene interactions on GDM susceptibility. Results 1. There were no significant differences between the two groups both in age( t= 0. 33, P = 0. 74) and pre-gestational BMI( t = 0. 45, P = 0. 33); 2. The distribution of genotype frequency in control group of the 2 studied SNPs conformed to HW equilibrium( P〉 0. 05). 3. The significant associations between polymorphisms of IGF2BP2rs1470579, PTPRD rs17584499 and GDMsusceptibility haven't been found in the dominant model, implicit model, co-dominant model, overdominant model and additive model. Conclusions The significant associations between 2 SNPs and GDM susceptibility haven't found in our study,but more samples were needed for further study.
