摘要
C18反相色谱柱是目前高效液相检测中,最常用到的一种色谱柱类型,在实际使用中由于样品性质或操作的原因,色谱柱会受到一定的损害。通过对常见的流动相误使用纯水而造成的疏水塌陷做分析,得到比较有效处理此类问题的方法。研究结果表明:在初期疏水塌陷发生后,经过甲醇-水体系先高比例有机相后低比例有机相梯度冲洗可以使柱效明显恢复,可以作为一种有效的挽救色谱柱的方法。
using C18 reversed phase chromatographic column is the highly effective liquid phase detection, one of the most commonly used to chromatographic column type, in be used actually because of sample quality or operation, chromatographic column will be damage.This article attempts through to the common mobile phase error caused by using pure water hydrophobic collapse for analysis, to get more effective ways to handle such problems.The results show that the hydrophobic collapse happens, after methanol - water system high organic phase first low organic phase gradient washing can make apparent recovery efficiency, can be used as an effective way to save the chromatographic column.
