具“异病同治”作用回乳抑增方组方优化及其量效关系分析 被引量:6

Formula optimization and dose-effect relationship of delactation and antiproliferation prescription based on treatment of different diseases with the same method

摘要 目的:以药效实验优化回乳抑增方,用量效关系解释其最佳药味组成。方法:利用高泌乳素血症大鼠模型,再次验证最佳组方对泌乳素、雌激素和孕激素的调节作用,应用HPLC法测定各拆方中6种药效成分含量,比较各拆方药理作用优劣与方中主要药效物质含量差异的关联。结果:与模型组比较,原方与优化方均可使泌乳素(PRL)明显降低,雌二醇(E2)、孕酮(P)显著升高,其中对PRL、E2、P的改变作用以优化方最为明显(P<0.01)。优化方中的玄参、浙贝母在不影响迷迭香酸含量的同时还能保证大麦芽碱的溶出,药味之间存在增效现象。而其他各组药味配伍均能影响君药中大麦芽碱和迷迭香酸的溶出。结论:优化方药味组成合理,方中主要药效物质含量较大,药效作用最佳。 OBJECTIVE To optimize the best formula composition of delactation and antiproliferation with efficacy experiment,and the dose-effect relationship was used to explain the optimal prescription.METHODS The hyperprolactinemia rat model was used to verify the regulation function of the best modified formula in prolactin,estrogen,progesterone level.Six components were detected by HPLC in each modified formula,and the relationship of bioactivity and the primary efficacy material was compared.RESULTS Compared to model group,PRL was obviously reduced,E2 and P significantly increased in original and optimized formulae,and the changes of PRL,E2 and P were most obvious in the optimized formula(P〈0.01).In the optimized formula,radix scrophulariae and zhejiang fritillary bulb increased hordenine dissolution and did not affect the content of rosemary acid,so there was a synergistic action between those herbs.In other modified formulae,dissolution of hordenine and rosemary acid were all influenced.CONCLUSION The optimized formula has a reasonable composition,higher content of major active ingredient,and optimal efficacy.
