摘要
目的建立柴胡中赪桐甾醇的含量测定方法,进行其资源调查。方法采用高效液相色谱法,Diamosil C18(2)(250mm×4.6mm,5μm)色谱柱;流动相为甲醇-乙腈(70-30);流速:1.0ml/min;检测波长:209nm;柱温:35℃。结果赪桐甾醇在1.60~205μg/ml范围内呈良好的线性关系(r=0.9999),平均回收率为100.99%,RSD=1.3%(n=9)。柴胡中赪桐甾醇的平均含量为0.385 mg/g,四川野生柴胡中赪桐甾醇含量最高。结论该方法简单准确,重复性好,可作为柴胡及其它含赪桐甾醇植物中赪桐甾醇质量评价方法。
Objective To establish an HPLC method to determine the content of clerosterol from different habitants.Methods The analysis was performed on Diamosil C18( 2)( 250 mm × 4. 6mm,5μm) column. The mobile phase consisted of methanol-acetonitrile( 70: 30) at a flow rate of 1. 0 m L/min with the gradient elution. The detection wavelength was 209 nm,the column temperature was 35℃. Results The standard curve of clerosterol showed a good linearity in the concentration range of 1. 6-205μg/m L and the average recovery was 100. 99%,and RSD value was 1. 3%( n = 9). Conclusion The content of clerosterol in Bupleuri Radix was 0. 385 mg/g,the highest content was wild from Sichuan. The established method is sample,accurate,and good repeatable. It could be used for the determination of clerosterol in Bupleuri Radix and other plants contained clerosterol.
