摘要 目的:探讨含半胱氨酸天冬氨酸蛋白水解酶3(Caspase-3)蛋白在人结直肠癌组织中的表达及其临床意义。方法:采用免疫组织化学SP法检测48例肠癌组织及配对癌旁组织中Caspase-3蛋白表达水平,并探讨其表达水平与临床病理学特征及预后的关系。结果:Caspase-3主要表达于胞浆,其在肠癌组织中的阳性表达率为27.08%,癌旁组织阳性率为58.33%,癌组织显著低于癌旁组织(P<0.05);Caspase-3阳性表达与肿瘤分期(P<0.05)和病理分级(P<0.05)有关。Caspase-3阳性患者中位无疾病进展生存时间为32.4个月,阴性患者为24.8个月(P>0.05)。结论:Caspase-3蛋白在人结直肠癌组织中表达率较低,可能结直肠癌的发生发展有关。 Objective To assess the clinical significance of Caspase-3 expression in colorectal cancer. Methods Forty eight patients with colorectal cancer were included in this study. The Caspase-3 expression was assayed by immunohistochemistry method for the cancer tissue and corresponding para-cancer tissue. The associations between Casepase-3 expression and clinical characteristics, prognosis were evaluated. Results Caspase-3 was mainly expressed in cytoplasm with the positive rate of 27.08% in cancer tissue and 58.33% in normal colorectal tissue. The positive rate in normal colorectal tissue was significant higher than that of cancer tissue with statistical difference(P〈0.05). Caspase-3 expression was associated with the tumor stage(P〈0.05) and pathology grading (P〈0.05). The disease free survival for positive and negative patients were 32.4 and 24.8 months with statistical difference HR=1.67, 95%CI:0.68-3.89(P〉0.05). Conclusion The expression rate of Caspase-3 protein in human colorectal cancer tissue is low, which may be related to the occurrence and develop-ment of colorectal cancer.
