摘要
目的:体外观察葫芦素E(CuE)联合顺铂(DDP)或紫杉醇(PTX)对非小细胞肺癌A549和H460细胞增殖的抑制作用。方法:采用MTT法检测CuE、DDP、PTX单独应用及CuE联合DDP或PTX对A549和H460细胞增殖的抑制作用。并采用CHOU-TALALAY联合指数法对药物联合应用的效果进行判定。结果:药物单用及CuE分别与DDP、PTX应用对A549和H460细胞的增殖均有抑制作用,药物合用时的中效浓度与单用时比较有不同程度的降低。CuE与DDP合用,对于A549细胞,表现为协同作用(CI<Ⅰ);对于H460细胞,大部分效应范围(0<fa<72%)为协同作用(CI<Ⅰ)。CuE与PTX合用,对于A549细胞,在较低效应范围(0<fa<14%)时为协同作用(CI<Ⅰ),在中高效应范围(14%<fa<100%)时则表现为拮抗作用(CI>Ⅰ);对于H460细胞,同样在较低效应范围(0<fa<28%)时为协同作用(CI<Ⅰ),在中高效应范围(28%<fa<100%)时则表现为拮抗作用(CI>Ⅰ)。结论:药物联合应用的效果在不同非小细胞肺癌细胞株之间存在差异,合理的联合用药可大大减少两种药物各自的用量,从而降低药物的毒副作用。
Objective To study the in vitro anti-proliferative effects of Cucurbitacin E (CuE) combined with cisplatin (DDP) or paclitaxel (PTX) in human non-small cell lung cancer cell lines A549 and H460. Methods MTT assay was performed to examine the growth inhibitory effects of CuE, DDP, PTX ,and CuE combined with DDP or PTX on A549 and H460 cells. And the effect of the combination therapy was evaluated by combination-index method of CHOU and TALALAY. Results Inhibitory effects were found when these 3 drugs were used alone or together. The median effect concentrations of these 3 drugs decreased to various extents when the drugs were used together. CuE was synergistic with DDP in A549 cells. In H460 cells, majority of effect range of CuE was synergistic with DDP. In A549 and H460 cells,low concentration of CuE was synergistic with low concentration of PTX, and middle-high concentration of CuE was antagonistic to middle-high concentration of PTX. Conclusion The effects of combination therapy are different in different human non-small cell lung cancer cell lines. Reasonable combination can greatly reduce the dosage of the two drugs, thus reducing the side effects.
