箍围药红肿消酊对皮肤脓肿大鼠血浆杀菌/通透性增加蛋白、消退素D1的影响 被引量:3

Effect of the Method of “Hoop Circumference” on Plasma Bactericidal/Permeability Increasing Protein and Resolvin D1 in Rats with Skin Abscess

摘要 目的:观察箍围药红肿消酊外用对皮肤脓肿大鼠血浆内杀菌/通透性增加蛋白(BPI)、消退素D1(RvD1)表达的影响。方法:将40只大鼠分为空白组、模型组、对照组、预治疗组、治疗组,除空白组外,其余4组皮下注射金黄色葡萄球菌造成皮下脓肿模型,模型组生理盐水纱条换药,对照组在注射细菌3 d后出现硬肿立即百多邦外涂换药,预治疗组在注射细菌后4 h立刻红肿消酊2 mL换药,治疗组在注射细菌3 d后出现硬肿立即红肿消酊2 mL换药,药物范围超过肿胀范围1 cm,1次/d。分别于治疗3 d、7 d、14 d、18 d心脏取血2 mL,ELISA法检测血浆内BPI、RvDl的表达。结果:除空白组外,其他用药4组血浆内BPI、RvDl含量均有不同程度的增高。在治疗3 d、7 d应用红肿消酊的预治疗组血浆内BPI含量升高明显,治疗7 d时,预治疗组与模型组相比升高明显,差别有统计学意义[(247±27.89)vs(225±13.75)ng/L,P<0.05];治疗14 d、18 d血浆内BPI表达量逐渐下降,预治疗组与对照组相比血浆内BPI表达量明显下降的势态,[(193±30.41),(183±26.95)vs(217±19.28),(205±23.89)ng/L,P<0.05],血浆内RvDl含量的表达在治疗过程中呈现不断增长势态,治疗14 d时,预治疗组[(2.11±0.24)ng/mL]、治疗组[(2.10±0.23)ng/mL]与模型组[(1.76±0.20)ng/mL]相比血浆内RvDl含量表达呈增高的势态(P<0.05);治疗18 d时,预治疗组[(2.17±0.27)ng/mL]、治疗组[(2.27±0.26)ng/mL]与模型组[(1.74±0.19)ng/mL]相比血浆内RvDl含量表达明显增高(P<0.01)。结论:应用红肿消酊换药治疗大鼠皮肤脓肿,可有效提高血浆内BPI、RvDl的含量水平,从而起到止痛、抗炎、抑制水肿等作用,促进护场形成,加速疮面愈合。 Objective To observe the effect of external use of encircling drugs Hongzhongxiao tincture on the expression of Bactericidal/permeability-increasing protein (BPI) and resolvin D1 (RvD1) in plasma of rats with skin abscess. Methods Forty rats were divided into blank group, model group, control group, pretreat-ment group and treatment group. Except for blank group, the remaining 4 groups were induced subcutaneous ab-scess by staphylococcus aureus subcutaneous injection. The model group was given saline gauze dressing, the control group was given mupirocin ointment dressing on 3 days after the injection of bacteria while hard swelling emerging, the pretreatment group was dressed with Hongzhongxiao tincture 2 mL at 4 hours after the injection of bacteria, the treatment group was dressed with Hongzhongxiao tincture 2 mL after 3 days of injection of bacteria. Drug range should be 1 cm more than the swelling range, once a day. At the treatment of 3 d, 7 d, 14 d, and 18 d, heart bloods (2 mL) were collected, and the contents of BPI and RvDl in plasma were detected using ELI-SA method. Results In addition to the blank group, the contents of BPI and RvDl in plasma was increased in other 4 groups. In the Hongzhongxiao tincture treatment at 3 d and 7 d, the contents of BPI increased obviously in the pretreatment group. In the treatment group at 7 d, the content of BPI was significantly higher than that in the model group, with statistically difference [(247±27.89) ng/L vs (225±13.75)ng/L,P〈0.05]. At 14 d and 18 d, plasma BPI content decreased gradually. Compared to the control group, the level of plasma BPI content in the pretreatment group was decreased significantly. The RvDl contents showed increasing tendency during the treatment period. At treatment of 14 d, the content of RvDl in pretreatment group [(2.11±0.24)ng/mL] and treat-ment group[(2.10±0.23)ng/mL] was higher that in the model group[(1.76±0.20)ng/mL] (P〈0.05).At treatment of 18 d, the content of RvDl in pretreatment group[(2.17 ± 0.27) ng/mL], treatment group[(2.27 ± 0.26) ng/mL] was significantly increased than that in model group [(1.74 ± 0.19) ng/mL]. Conclusion The treatment of rat skin abscess with Hongzhongxiao tincture can effectively improve the contents of BPI and RvDl in plasma. Thus, it plays the role of analgesia, anti-inflammatory, inhibiting edema, and, promote the formation of the protective field, accelerate the healing of sores.
