一株苯酚降解菌分离及降解特性研究 被引量:3

摘要 从实验室模拟煤化废水处理装置的好氧段取污泥,先利用含苯酚的富集培养基筛选出对苯酚具有良好耐受性能的菌株(记为JX,JY,JZ);再利用以苯酚为唯一碳源的无机盐培养基,从以上三种菌株中筛选出一株对苯酚具有良好降解性能的菌株(JY)。对JX,JY,JZ的培养特征以及生理生化特征进行观察与鉴定。将菌株JY进行了16S r DNA基因序列的测定,最终确定该菌株为不动杆菌属(Acinetobacter)。并研究了菌投加量、温度、p H、以及摇床转速对JY的生长量以及酚降解率的影响。结果表明:菌株JY的最佳菌投加量为3%,最适温度为30℃~35℃,最佳p H为7.0,最佳摇床转速为120 r/min。 The sludge is taken from the experimental aerobic stage of the simulated coal chemical wastewater treatment plant. Firstly, strains with good tolerance to phenol (JX, JY, JZ) were screened out by enrichment medium containing phenol. Then, a strain (JY) with good degrading ability to phenol was screened from the a- bove three strains by using the inorganic salt medium with phenol as sole carbon source. The culture character- istics and physiological and biochemical characteristics of JX, JY and JZ were observed and identified. The strain JY was determined by 16S rDNA gene sequence. Finally, the strain was identified as Acinetobacter. The effects of the dosage of bacteria, temperature, PH, and the rotation speed on the growth of JY and the degrada- tion rate of phenol were studied. The results showed that the optimal dosage of JY was 3 %, the optimum tem- perature was 3℃-35 ℃,the optimum pH was 7.0, and the optimum rotational speed was 120r/min.
