摘要
目的:Numb蛋白在乳腺癌中有不同程度的表达缺失,本研究旨在探讨乳腺癌组织Numb蛋白的表达,及其与表皮生长因子受体2(HER-2)、雌激素受体(ER)、孕激素受体(PR)表达的关系。方法:收集2015年8月-2017年3月江西省南昌市第三医院收治的200例乳腺癌患者的临床病理资料,同时应用免疫组织化学Elivison方法检测200例乳腺癌组织的Numb、HER-2、ER和PR的表达。结果:Numb蛋白在乳腺癌组织中的阳性表达率51.0%(102/200),淋巴结有转移者Numb表达高于淋巴无转移者(x^2=6.72,P=0.02),HER-2阳性者Numb表达低于HER2阴性者(x^2=7.00,P=0.02),ER阳性者Numb表达高于ER阴性者(x^2=13.55,P=0.00)。Luminal型Numb阳性表达率显著高于HER-2过表达型和三阴型(x^2=7.26,P=0.01),HER-2过表达型和三阴型Numb阳性表达率比较差异无统计学意义(x^2=0.76,P=0.12)。Numb表达在不同年龄、肿瘤大小、组织学分级及PR状态间差异均无统计学意义(x^2=0.13,P=0.72;x^2=0.55,P=0.46;x^2=0.46,P=0.59;x^2=0.06,P=0.80)。结论:Numb蛋白在乳腺癌中有较高表达,其表达与HER-2、ER和淋巴结转移有关,Numb能否成为乳腺癌新的治疗靶点或预测因子有待进一步研究。
Objective: The expression of Numb protein in breast carcinomas had different degrees of expression, to explore the expression of Numb protein in breast carcinomas and examine the correlation between its expression and expression of human epidermal growth factor receptor-2 ( HER-2 ), estrogen receptor ( ER ),ogesterone receptor ( PR ) .Method: The clinicopathological data of' 200 breast cancer patients admitted in the Third Hospital of Nanchang from August 2015 to March 2017 were collected and analyzed.Immunohistochemical method was used to detect the expression of HER-2, ER, PR and Numb protein.Result: The rate of Numb protein expression was 51.0% ( 102/200 ), the expressions of Numb in ER positive patients were significantly higher than those of ER negative patients, and this trend extended to HER-2 negative and no node metastasis patients ( X^2=13.55, P=0.00; X^2=7.00, P=0.02; X^2=6.72, P=0.02) . The expressions of Numb protein in Luminal subtype was signifycantly higher than in HER-2 over-expression subtype and basal like subtype ( X^2=7.26, P=0.01 ), but there was no significant difference between HER-2 over-expression subtype and basal like subtype ( X^2=0.76, P=0.12 ) .No correlations existed between the expression of Numb protein and age, tumor size, histologic grade or PR of breast cancer patients (X^2=0.13, P=0.72;X^2=0.55, P=0.46;X2-0.46, P=0.59;X^2=0.06, P=0.80) . Conclusion: Numb protein is highly expressed in breast cancer, and its expression is related to lymph node status, HER-2 and ER, and node metastasis.However, Numb can be a new therapeutic target or predictor for breast cancer remains to be further studied.
