摘要 目的探讨两种盐酸脱钙液对骨髓组织染色质量的影响。方法选取首都医科大学附属北京同仁医院的骨髓穿刺标本299例,在常温下分别采用8%盐酸甲醛脱钙175例(实验组),30%盐酸甲醛脱钙124例(对照组),比较两组的脱钙时间、苏木素-伊红(HE)染色效果。另从两组中分别选取正常骨髓标本36例,以免疫组化Envision法染色,观察髓过氧化物酶(myeloperoxidase,MPO)、CD61、CD235a及Ki-67抗原表达情况。结果对照组的脱钙时间为(50.42±13.91)分钟,实验组的脱钙时间为(66.83±17.59)分钟,经比较差异无显著性(P>0.05),且HE染色效果均佳;MPO和CD235a在两组中表达差异性不大,CD61在实验组中高表达率为97.2%(35/36),在对照组中高表达率为83.3%(30/36),差异有显著性(P<0.05);Ki-67在实验组中高表达率为97.2%(35/36),在对照组中高表达率为8.3%(3/36),差异有显著性(P<0.05)。结论 8%盐酸甲醛脱钙液脱钙后HE及免疫组化染色效果均佳,能很好地满足临床骨髓组织活检的脱钙需要。 Objective To investigate the influences of two different decalcified hydrochloric acid on the quality of the bone marrow biopsy. Method 299 bone marrow biopsy specimens were selected, and divided into two groups(experimental group and control group). 8% hydrochloric acid formaldehyde fluid were used into experimental group with 175 cases and 30% hydrochloric acid formaldehyde liquid were used into control group with 124 cases at the normal temperature respectively. Then the time of decalcification, HE staining of two groups were comparaed and analyzed.In addition, 36 cases of normal bone marrow were selected separately for immunohisto-chemical staining, The expression of MPO, CD61, CD235a and Ki-67 was observed by immunohistochemistry Envision method. Result The decalcification time of the control group was (50.42&#177;13.91)min, the experimental group was (66.83&#177;17.59)min, and there was no significant difference in statistical analysis, but HE staining were good; MPO and CD235a were not different in the two groups, CD61 had a high expression rate of 97.2% (35/36) in the experimental group and 83.3% (30/36) in the control group. There was significant difference between the two groups (P〈0.05); Ki67 had a high expression rate of 94.4% (34/36) in the experimental group and 8.3% (3/36) in the control group. There was significant difference between the two groups (P〈0.05). Conclusion HE staining and immunohistochemical antigen expression of 8% hydrochloric acid formaldehyde decalcification are good, can well meet the needs of clinical bone marrow biopsy decalcification.
