体外模型在药源性肾毒性评价中的研究进展 被引量:4

摘要 由于肾毒性物质的靶器官选择性,药物诱导的肾毒性是目前新药开发和临床应用中存在的重要问题,高通量肾毒性体外评价模型可以提高大量化合物的筛选效率,节约时间和成本,已经逐渐成为肾毒性研究的主要研究方法。虽然肾毒性体外评价模型的研究热度不断升温,但由于肾毒性的特异性,仍未能建立标准的体外评价模型。目前,肾毒性体外评价的研究主要基于传统的2D细胞模型,3D细胞模型尚在初级阶段。本文对近年来高通量肾毒性体外评价模型的研究进展做一综述。 The kidney is a major target for drug-induced toxicity. Drug-induced nephrotoxicity remains a major problem in research development of drugs and clinical settings. The high throughput screening( HTS) in vitro model has become an important research method,which can improve screening efficiency of many compounds in time and cost. Though there are more and more researches on in vitro models for assessment of drug-induced nephrotoxicity,validated nephrotoxic prediction model is still absent because of nephrotoxicity specificity. At present,the nephrotoxicity research is mainly based on traditional 2D cell model,and 3D cell model is at the stage of exploration. This review summarizes the latest advances of HTS in vitro nephrotoxicity assessment models.
