血根碱对小鼠肝细胞色素P450与b5的影响

摘要 研究血根碱对小鼠肝细胞色素P450与b5的影响,为血根碱的兽医临床应用提供实验依据。将血根碱按高剂量[21 mg/(kg·d)]、中剂量[10.5 mg/(kg·d)]、低剂量[2.1 mg/(kg·d)]给小鼠给连续灌胃给药7 d后,采用考马斯亮蓝法测定小鼠肝微粒体蛋白含量;紫外-可见光分光光度法测定细胞色素P450和细胞色素b5的含量。结果显示,与空白组相比,高、中剂量组小鼠肝微粒体蛋白含量、细胞色素P450和b5活性降低(P<0.05),而低剂量组无显著变化(P>0.05)。结果表明,高、中剂量组血根碱对小鼠肝细胞色素P450和b5有抑制作用。 In order to provide experimental basis for the clinical application of sanguinarine, its effect on mice hepatic cytochrome P450 and b5 was studied. Three treatment groups and control group were divided up. Mice in high-dose, middle-dose and low-dose groups were respectively administrated sanguinarine at 21 mg/(kg·d), 10.5 mg/(kg·d) and 2.1 mg/(kg·d) orally for 7consecutive days. The bradford method was used to determine the protein concentration of hepatic microsomes, and the activities of cytochrome P450 and b5 were determined by visible-ultraviolet spectrophotometer. The results showed that the liver microsomal protein content, and cytochrome P450 and b5 activities were decreased in high-dose, middle-dose groups compared with the blank group(P0.05), while the low-dose group had no significant change(P0.05). The results indicated that high-dose and middledose of sanguinarine could inhibit mice cytochrome P450 and b5 activities.
