摘要
目的鉴定白扁豆及其混伪品ITS2条形码,为白扁豆药材鉴定提供新方法。方法采用PCR法扩增ITS2序列,并双向测序,经Codon Code Aligner拼接后,利用MEGA软件进行数据处理,构建系统聚类树(NJ树)。结果白扁豆ITS2序列长度为218 bp,由所构建的系统聚类树图可见,各物种均表现单系性,同时又可与其它物种明显区分。结论 ITS2条形码适用于白扁豆及其混淆品的快速鉴别,进一步验证了DNA条形码技术应用于中药材鉴别的有效性。
Objective To investigate the applicabsility of ITS2 barcoding in discriminating Lablab Semen Album and its adulterants. Methods The internal transcribed spacer 2 (ITS2) regions were amplified by PCR and sequenced bi- directionally. Then the obtained sequences were assembled using the CodonCode Aligner. Identification analyses were performed using neighbor-joining (NJ) methods. Results The length of ITS2 sequence of Lablab Semen Album was 218 bp. In the cluster dendrogram, all species showed monophyletic, and distinguished from others. Conclusion ITS2 sequence can be used as DNA barcoding to identify Lablab Semen Album and its adulterants effectively and our study can further confirm the effectiveness of ITS2 to identify traditional Chinese medicinal materials.
