玉米须总皂苷对非酒精性脂肪肝病大鼠肝脏PPARγ表达的影响 被引量:7

摘要 目的探讨玉米须总皂苷(ZMLS)对非酒精性脂肪肝病(NAFLD)大鼠的治疗作用及其作用机理。方法同一批次SD大鼠(60只)在适应性喂养10 d后随机分为正常组,模型组以及玉米须总皂苷治疗组(n=20),造模成功后,正常组和模型组给予1 g/(kg.d)生理盐水灌胃,玉米须总皂苷治疗组给予4 mg/(kg.d)灌胃治疗,分别于治疗4、8周后处死大鼠各半,检测相关生化治疗和肝脏中过氧化物酶体增殖物激活受体γ(PPARγ)表达。结果与正常组相比,模型组AST、ALT、IL-1、IL-6、FBG、TG、FFA明显升高(P<0.05),FINS明显降低(P<0.05),治疗4周后AST、ALT、IL-1、IL-6、FBG、TG、FFA明显降低(P<0.05),FINS明显升高(P<0.05);治疗8周后AST、ALT、IL-1、IL-6、FBG、TG、FFA进一步降低(P<0.05),FINS进一步升高(P<0.05);免疫组化、RT-PCR以及Western Blot可见模型组PPARγ表达明显降低(P<0.05),治疗4周后有明显升高(P<0.05),治疗8周进一步升高。结论玉米须总皂苷对非酒精性脂肪肝病肝功能有明显的调节作用,可能与其调节PPARγ表达,进而改善炎症反应,降低血脂有关。 Objective To investigate whether Zea Mays L(ZMLS) has therapeutic effect and its mechanism of liver in non alcohol fatty liver disease(NAFLD) rats. Methods After feeding adaptation 10 d, the same batch SD rats(60) were randomly divided into normal group, model group and ZMLS treatment group(n=20), after successful built non-alcoholic fatty liver disease model, the normal group and model group were given 1 g/kg/d normal saline, ZMLS group was received 4 mg/(kg.d)orally, after treatment 4 and 8 weeks, the rats were sacrificed and detected biochemical and liver PPARγ expression. Results Compared with normal group, the model group in AST, ALT, IL-1, IL-6, FBG, TG and FFA were increased significantly(P〈0.05), FINS was decreased significantly(P〈0.05), after 4 weeks of treatment, AST, ALT, IL-1, IL-6, FBG, TG and FFA were decreased(P〈0.05), FINS was increased significantly(P〈0.05); After 8 weeks of treatment, AST, ALT, IL-1, IL-6, FBG, TG and FFA were further reduced(P〈0.05), FINS was further increased(P〈0.05); immunohistochemistry, RT-PCR and Western Blot can seen the expression of PPARγ in model group was decreased significantly(P〈0.05), after 4 and 8 weeks of treatment was significantly higher the model group(P〈0.05), 8 weeks of treatment further increased. Conclusion To non-alcoholic fatty liver disease, ZMLS can significantly improve its liver function, which may be related to this drug can regulated the expression of PPARγ, thereby regulate the inflammation, improve blood lipid related.
