烟草黄斑驳坏死病毒的提纯及抗血清制备

PURIFICATION AND ANTISERA PREPARATION OF TOBACCO YELLOW MOTTLE NECROSIS VIRUS

烟草黄斑驳坏死病毒(TYMNV)是近几年来从我国烟草上新分离到的一种病毒,引起烟草植株的黄斑驳坏死症状。本文在分离纯化病毒的基础上,用差速离心法部分提纯了该病毒,其提纯产量约为20mg/公斤病叶。该病毒的免疫原性较弱,采用从次肌肉注射乳化病毒免疫家兔,可制备抗血清。用 SDS—琼脂免疫双扩散法测定,其抗血清效价为1/16。该血清特异性较好,可用于 TYMNV 的田间检测和血清学关系的研究。

Tobacco Yellow Mottle Necrcrsis Virus(TYMNV)is a newly isolated virus from the tobacco fields in China.It causes yellow mottle necrosis on tobacco plant.A procedure for the partial purification of TYM- NV was developed and the purification yield was about 20mg per Kg diseased tobacco leaves.The antisera of this virus has been produced by injecting the rabbits intramusculary with the partial purified TYMNV. The immunogenicity of the virus is poor and give the antisera with liters of 1/16 in SDS-gel-diffusion tests.The antisera is specific and can be used in detecting TYMNV in tobacco fields.
