摘要
目的观察以一种主要成分为苄索氯铵和异丙醇的口腔印模专用消毒剂对口腔印模表面乙肝病毒活性及免疫原性的影响。方法使用消毒液对HBV污染的口腔印模进行喷雾消毒,检测HBs Ag含量和HBV-DNA含量,评价消毒效果;检测消毒前后HBV在体外诱导淋巴细胞分泌IFN-γ和IL-2含量,评价对HBV免疫能力的影响。结果喷雾消毒3 min和15 min,对HBs Ag和HBV-DNA的灭活效果明显高于消毒1 min组,灭活率分别为(98.8±1.2)%、(98.9±1.1)%和(99.3±1.1)%、(99.4±1.3)%。消毒3 min和15 min HBV诱导淋巴细胞IFN-γ相对表达量明显高于空白对照和未消毒组;与空白细胞组比较,未消毒组HBV诱导淋巴细胞IL-2相对表达量明显增加,而各消毒组的IL-2相对表达量未发生显著变化。结论该消毒剂喷涂口腔印模时,对HBV的最佳消毒时间为3 min。此时HBV仍然保持较强的免疫原性,能够诱导淋巴细胞IFN-γ表达水平上调,对IL-2表达水平无影响。
Objective To explore the effect of the compound disinfectant used specially for the oral impression disinfection on hepatitis B virus( HBV) activity and immunogenicity. Methods After the oral impression polluted by HBV being sprayed with Cavicide disinfector,the disinfection effect was evaluated with the levels of HBV surface antigen( HBs Ag) and HBV DNA. At the same time,the effect of this disinfectant on HBV immunity ability was assessed by detection of the expression levels of IFN-gamma and IL-2 in lymphocytes induced with HBV induction in vitro. Results In the group of Cavicide disinfection for 3 min and 15 min,the inactivated efficiency for HBs Ag and HBV-DNA were( 98. 8 + 1. 2) % 、( 98. 9 + 1. 1) % and( 99. 3 ± 1. 1) % 、( 99. 4 ± 1. 3) %,which were significantly higher than that of Cavicide disinfection for 1 min group( P〈 0. 01). Compared with blank cell group and with no disinfection group,that in the group of Cavicide disinfection for 3 min and 15 min,the levels of IFN-gamma significantly increased in lymphocytes by HBV induction( P〈 0. 01). Conclusion When disinfecting the oral impression with this compound disinfectant,the appropriate action time is 3min for HBV inactivation. After the oral impression disinfected with Cavicide for 3 min,HBV remain strong immunogenicity,which can upregulate the IFN-gamma expression,but have no effect on the IL-2 expression.
