摘要
[目的]探索软枣猕猴桃"寒玉1号"适宜组培方法。[方法]以新生腋芽为外植体,筛选最佳消毒方法,并对影响其生长的NAA、6-BA等生长调节物质的浓度进行筛选。[结果]以5%NaClO 8 min+0.1%HgCl_26 min或9 min,5%NaClO 10 min+0.1%HgCl_26 min为适宜消毒方法,污染率和褐化率均较低;腋芽在WPM+6-BA 3.0 mg/L+NAA 0.2 mg/L培养基中增殖效果最佳,增殖系数可达5.6,株高3.9 cm;丛生芽在WPM+NAA 0.3 mg/L培养基中诱导生根效果最佳,生根率可达100%,平均根条数为4.4条,平均根长为5.2 cm。[结论]软枣猕猴桃"寒玉1号"可采用组织培养的方法进行快速繁殖。
[ Objective ] To explore the suitable tissue culture technique for Actinidia arguta "Hanyu No. 1 ". [ Method ] With axillary bud as explant, the best disinfection method was filtered, and the NAA, 6-BA concentration were selected. [ Result ] The best disinfection methods were 5% NaClO 8 min + 0.1% HgCl2 6 min or 9 min, 5% NaClO 10 min + 0.1% HgCl2 6 min, in which the mortality rate and contamination rate were lower. The best medium of inducing geiminating of expiant was WPM + 6-BA 3.0 mg/L + NAA 0.2 mg/L, in which the ratio of proliferation was 5.6 and the height of plant was 3.9 cm. The best medium of inducing rooting from cluster buds was WPM + NAA 0.3 mg/L, in which the rate of rooting was 100% , the average number of root was 4.4 and the average length of root was 5.2 cm. [ Conclusion] It concluded that Actinidia arguta "Hanyu No. 1" can adopt the method of tissue culture for rapid propagation.
