摘要
以美国红枫十月辉煌带腋芽茎段为外植体,以不同植物激素类型及浓度配比对其腋芽启动、丛芽增殖和生根等阶段进行试验研究。结果表明,腋芽在诱导培养基MS+TDZ 0.008 mg/L+BA 0.1 mg/L上的诱导率达80%以上。丛芽增殖的最优培养基为MS+TDZ 0.005 mg/L+BA 0.05 mg/L+GA31.5 mg/L+琼脂粉6.5 g/L+蔗糖30 g/L,增殖倍率达3.58。生根培养基1/2MS+IBA 0.8 mg/L+NAA 0.5 mg/L+琼脂粉6.5 g/L+蔗糖20 g/L,获得90%以上的生根率,根数2~5条。
The stem segments with axillary buds ofAcer rubrum 'ctober Glory' were taken as explants to culture by different hormone types and concentration ratios, doing a series of experiments about buds induction and proliferation ,rooting and so on.The best induction medium was MS+TDZ 0.008 mg/L+BA 0.1 mg/L+agar 6.5 mg/L+sugar 30 g/L,the induction rate was above 80% ,the optimum multiplication medium was MS+TDZ 0.005 mg/L+ BA 0.05 mg/L+GA3 1.5 mg/L+agar 6.5 mg/L+sugar 30 g/L ,the proliferation multiple was 3.58.The best rooting medium was 1/2MS+IBA 0.8 mg/L+NAA 0.5 mg/L+agar 6.5 mg/L+sugar 20 g/L,rooting rate reached 90% ,root number was 2-5.
