安胃汤干预慢性萎缩性胃炎大鼠JAK/STAT信号传导通路的作用机制 被引量:14

摘要 目的:从JAK/STAT信号传导通路角度探讨安胃汤对慢性萎缩性胃炎模型大鼠疗效的作用机制。方法:采用甲基硝基亚硝基胍(MNNG)慢性萎缩性胃炎大鼠模型,应用安胃汤进行干预;采用实时荧光定量PCR方法、Western blot检测JAK1、STAT3、SOCS-3、c-myc基因表达及其蛋白含量的水平。结果:该方可以明显下调JAK1、STAT3、c-myc基因以及其蛋白含量的表达,升高SOCS-3基因以及其蛋白含量的表达。结论:安胃汤可能通过调节JAK1/STAT3信号传导通路关键因子JAK1、STAT3、c-myc、SOCS-3的表达,缓解炎症并防治慢性萎缩性胃炎。 Objective: To explore the mechanism of Anwei Decoction on model rats with chronic atrophic gastritis(CAG) from the perspective of the AK/STAT signaling pathway. Methods: MNNG was used to establish models of CAG rats, which were then subject to the intervention by Anwei Decoction; Quantitative real-time fluorescent PCR and Western blot were used to measure the expression of genes JAK1, STAT3, SOCS-3 and cmyc and their protein levels. Results: The formula significantly down-regulated the expression of genes JAK1,STAT3 and c-myc and their protein levels, and up-regulated the expression of gene SOCS-3 and its protein level. Conclusion: Anwei Decoction may relieve inflammation and inhibit atrophic gastritis by regulating the expression of key factors JAK1, STAT3, c-myc, and SOCS-3 in the JAK1/STAT3 signaling pathway.
