摘要
目的:采用响应面法优化新疆黑果小檗根皮中总生物碱提取工艺。方法:以总生物碱提取率为考察指标,通过单因素试验,确定提取次数为3次,提取温度:80℃。采用响应面法优化乙醇浓度、提取时间、料液比3个因素。结果:得到最佳提取工艺为:乙醇体积分数50%,料液比1:51(g·m L^(-1)),提取时间2 h,提取次数3次,提取温度80℃。在该条件下,平行3次验证实验,总生物碱提取率为4.01%(RSD%=1.12%),可达到预测值的99.01%。结论:采用响应面法优化的黑果小檗根皮中总生物碱提取工艺稳定可行。
Objective: To optimize the extraction process of total alkaloids from Xinjiang Berberis Heteropoda root bark. Methods: The content of total alkaloids was determined by Acid-Dyes-UV Spectrophotometry. The optimal process parameters were ethanol at 80 ℃ and three extraction cycles. With the content of total alkaloids as index, based on single factor experiments, the ethanol concentration, extraction time and solid-liquid ratio as influence factors, three factors and three levels Box-Behnken design of Response Surface Methodology(RSM)was adopted. Results: The optimal process parameters were as follows: extracted 2 h with 51 times 50% ethanol at 80 ℃, three extraction cycles. Under this condition, three paralle tests were carried out, yeild of the total alkaloids could reach 4.01%, RSD%=1.12%, which accounted for 99.01% of the predicted value. Conclusion: The optimized extraction process is stable and feasible, to provide a basis for subsequent separation and purification of the total alkaloids from Berberis Heteropoda root bark.
