LncRNA RAB11B-AS1在胃癌组织中的表达及临床意义

Expression and clinical significance of LncRNA RAB11B-AS1 in gastric cancer

目的探讨长链非编码(LncRNA)RAB11B-AS1在胃癌组织中的表达及其临床意义。方法通过实时荧光定量PCR(QRT-PCR)检测LncRNA RAB11B-AS1在83例胃癌组织及正常胃组织标本中的表达,分析其表达与患者临床病理特征及预后的关系。结果 LncRNA RAB11B-AS1在胃癌组织中的相对表达量为6.954±1.080,高于正常胃组织的1.061±0.090,差异有统计学意义(P<0.05)。LncRNA RAB11B-AS1表达与性别、年龄及肿瘤部位无关(P>0.05);而与临床分期、淋巴结转移、远处转移、分化程度及生存状态有关(P<0.05)。LncRNA RAB11B-AS1高表达者的中位无进展生存期(PFS)和中位总生存期(OS)分别为19.0个月和29.0个月,低于低表达者的32.0个月和43.0个月,差异均有统计学意义(P<0.05)。Cox多因素回归分析提示,LncRNA RAB11B-AS1表达、远处转移及临床分期是胃癌独立的预后因素(P<0.05)。结论LncRNA RAB11B-AS1参与调节胃癌的发生发展,可作为胃癌诊断和预后评估的潜在分子标志物。

Objective To investigate the expression and clinical significance of long non-coding RNA（ LncRNA） RAB11BAS1 in gastric cancer. Methods The expression of LncRNA RAB11B-AS1 was detected by real-time quantitative PCR（ QRT-PCR） in83 cases of gastric cancer tissues and normal gastric tissues. The relationship of LncRNA RAB11B-AS1 expression with the clinicopathological features and prognosis of the patients was analyzed. Results The expression of LncRNA RAB11B-AS1 in gastric carcinoma tissue was 6. 954 ± 1. 080,higher than 1. 061 ± 0. 090 in normal gastric tissue,and the difference was statistically significant（ P〈0. 05）. The expression of LncRNARAB11B-AS1 was not related to sex,age and tumor location（ P〉0. 05）,but related to clinical stage,lymph node metastasis,distant metastasis,tumor differentiation and survival status（ P〈0. 05）. The median progression free survival（ PFS） and overall survival（ OS） of patients with high expression of LncRNA RAB11B-AS1 was 19. 0 months and 29. 0months respectively,which was lower than 32. 0 months and 43. 0 months of patients with low expression of LncRNA RAB11B-AS1（ P〈0. 05）. Cox multivariate regression analysis suggested that LncRNA RAB11B-AS1 expression,distant metastasis and clinical stage were independent prognostic factors of gastric cancer（ P〈0. 05）. Conclusion LncRNA RAB11B-AS1 is involved in the regulation of gastric cancer,and can be used as a potential molecular marker for the diagnosis and prognosis of gastric cancer.
