摘要
为了研究猪繁殖性状候选基因骨桥蛋白(osteopontin,OPN)和视黄醇结合蛋白4(Retinol-binding proteins,RBP4)的多态性在大白猪群体中的遗传效应,采用PCR和PCR-RFLP技术在大白猪群体中对与繁殖性能相关的候选基因OPN和RBP4的基因多态性进行检测,并对不同基因型的总产仔数、产活仔数及健仔数进行关联性分析。结果显示:OPN和RBP4基因的优势基因都是B等位基因,在初产母猪中OPN基因AA型比BB型个体在总产仔数、产活仔数及健仔数上分别高0.80、0.22和0.28头(P>0.05);RBP4基因AA型比BB型个体分别高0.11、0.46、0.29头(P>0.05)。在经产母猪中OPN基因AA型比BB型个体总产仔数高0.05头(P>0.05),而产活仔数和健仔数分别低0.10头(P>0.05)和0.22头(P<0.05);RBP4基因BB型比AA型个体在总产仔数、产活仔数和健仔数分别高0.12、0.08和0.02头(P>0.05)。在合并基因型中,初产母猪合并基因型总产仔数和产活仔数最高的ABAB型比最低的BBAB型个体多3.26头(P<0.05)和3.16头(P<0.05),健仔数最高ABAB型比最低ABBB型个体多3.08头(P<0.05);经产母猪合并基因型在总产仔数、产活仔数和健仔数最高ABAA型比最低AAAA型个体多0.67(P>0.05)、1.1(P>0.05)和1.45头(P<0.05)。本试验结果可为猪分子标记辅助育种提供依据。
The present study was conducted to examine the genetic effects of the OPN and RBP4 genes on reproductive traits in Yorkshire sows. The polymorphism of OPN and RBP4 genes were detected by PCR,PCR-RFLP and electrophoresis,and the correlative analysis of genotypes and reproductive traits was made. The results showed that allele B was the dominant allele in both OPN and RBP4 genes. In the first farrowing sows,the total number born( TNB),number born alive( NBA) and number healthy birth( NHB) of genotype AA were 0. 80,0. 22 and 0. 28 for OPN gene,respectively,higher than that of genotype BB( P〉0. 05). For RBP4 gene,the TNB,NBA and NHB of genotype AA were 0. 11,0. 46 and 0. 29,respectively,higher than that of genotype BB( P〉0. 05). In multiparous sows,for OPN gene,the TNB of genotype AA was 0. 05 higher than that of genotype BB,but the NBA and NHB were 0. 1( P〉0. 05) and 0. 22( P〈0. 05) lower;for RBP4 gene,the TNB,NBA and NHB of genotype BB were 0. 12,0. 08 and 0. 02 higher than that of genotype AA( P〉0. 05),respectively. In the combined genetypes,in the first farrowing sows,the TNB and NBA of genotype ABAB were 3. 26( P〈0. 05) and 3. 16( P〈0. 05) higher than genotype BBAB,the NHB of genotype ABAB was 3. 08( P〈0. 05) higher than that of genotype ABBB. In the multiparous sows,the TNB,NBA and NHB of genotype ABAA were 0. 67( P〉0. 05),1. 10( P〉0. 05) and 1. 45( P〈0. 05) higher than that of genotype AAAA. Our finding can provide the basis for molecular marker assisted breeding in pigs.
