摘要
为了对缅甸98谱系O型口蹄疫病毒不同宿主适应毒的3A编码基因序列及其氨基酸序列进行分析,从而研究O型口蹄疫病毒的演化、变异情况,将O型口蹄疫病毒株O/XJ/10-11接种于牛、乳鼠、猪及BHK-21细胞,获得相应的适应毒,以各组织液中的口蹄疫病毒RNA为模板,反转录并扩增3A基因,采用Vector NTI 11.5和DNA Star生物学软件对3A基因进行比对分析。结果表明:牛、乳鼠、猪、BHK-21细胞4种宿主适应毒,3A基因较稳定,变异较少;变异均发生在3A的第99、114位氨基酸上,说明FMDV 3A基因的第99、114位氨基酸在一定程度上与宿主嗜性有关。本试验结果为进一步分析O型口蹄疫病毒不同宿主适应毒基因组的变异奠定了基础。
In order to analyze the nucleotide and amino acid sequences of the gene encoding 3A protein of Mya98 lineage foot and mouth disease virus( FMDV) type O adapted in different hosts and therefore viral evolution and variation of FMDV,FMDV strain O/XJ/10-11 was inoculated in bovine,swine,rat and BHK-21 cells and the host-adapted FMDV strains were obtained. RNA in tissue fluid was extracted and used as the template for reverse transcription and amplification of 3A gene. Then these sequences were analyzed by Vector NTI 11. 5 and DNAStar biological software. The results showed that in the four host-adapted virus strains from bovine,rat,swine and BHK-21 cells,3A gene was more stable,and variation occurred in aa 99 and aa 114,indicating that the two amino acids of FMDV 3A gene are related to host tropism to a certain extent. The results lay a foundation for further analysis of the variability of the genomes from the different host-adapted FMDV strains.
