白藜芦醇对阿尔茨海默病小鼠认知功能及脑皮质Aβ_(1-40)和Aβ_(1-42)表达的影响 被引量:6

摘要 目的观察白藜芦醇(Res)对阿尔茨海默病(AD)小鼠认知功能及脑皮质β淀粉样蛋白(Aβ_(1-40))和Aβ_(1-42)的影响。方法将30只雌性昆明小鼠编号后采用随机数字表法分为对照组、模型组和Res组,每组10只。采用120 mg/kg D-半乳糖联合10 mg/kg氯化铝构建AD小鼠模型,连续给药90 d。对照组腹腔注射和灌胃等体积生理盐水。Res组造模第31天每天灌胃44 mg/kg Res,连续灌胃60 d。其余各组给予等体积生理盐水灌胃。采用跳台和Morris水迷宫实验测试小鼠认知功能,免疫组织化学检测小鼠脑皮质Aβ_(1-40)和Aβ_(1-42)表达。结果与模型组小鼠比较,Res组小鼠跳台学习成绩明显提高、潜伏期明显延长、错误次数明显减少,Morris水迷宫逃避潜伏期明显缩短、目的象限停留时间和穿环次数明显增多,脑皮质Aβ_(1-40)和Aβ_(1-42)表达明显减少,差异均有统计学意义(P<0.05)。结论 Res对AD小鼠认知功能具有改善作用,其机制可能与降低脑内Aβ_(1-40)和Aβ_(1-42)表达有关。 Objective To study the effect of resveratrol( Res) on cognitive function and the expression of Aβ_(1-40) and Aβ_(1-42) in cerebral cortex of model mice with Alzheimer′s disease(AD). Methods Kunming female mice were divided into the control group, the model group and the Res group according to the random number table,10 mice in each group. AD model mice were established by D-galactose(120 mg/kg) combining with AlC l3(10 mg/kg) for 90 days. Same volume of saline were given to mice in the control group by intraperitoneal injection and gastric irrigation. at 31 d ays after modeling,The Res group was treated with Res by gavage at doses of 44 mg/kg a day for 60 days.The rest groups were intervented with same volume of saline by intramus-cular injection,the cognitive function was tested by the step-down test and Morris water maze test. The expression of Aβ_(1-40) and Aβ_(1-42) in cerebral cortex were measured by immunohistochemistry. Results Compared with the model group,the achievement of step down test in the Res group was increase,the latent period was longer,the number of mistakes was decreased;the escape latency was shorter,the target quadrant time and the number of quadrant entries were more,the expression of Aβ_(1-40) and Aβ_(1-42) in cerebral cortex were decreased,the difference was stastically significant(P0.05). Conclusion Res may improve the cognitive function of AD model mice,which is associated with decreased expression of Aβ_(1-40) and Aβ_(1-42).
