1,25-二羟基维生素D_3干预前后糖尿病肾病患者外周血单核细胞促炎细胞因子的表达情况 被引量:2

摘要 目的探讨1,25-二羟基维生素D3(VD3)干预前后糖尿病肾病患者外周血单核细胞促炎细胞因子的表达情况。方法纳入2型糖尿病合并糖尿病肾病(DN)患者(DN组)及健康对照者(对照组)各15例,分别检测两组外周血单核细胞培养上清液在VD3干预和非干预条件下白介素(IL)-17A、干扰素(IFN)-γ、肿瘤坏死因子(TNF)及IL-2水平。结果VD3干预前,两组IL-17A水平比较差异无统计学意义(P>0.05),DN组IFN-γ、TNF和IL-2水平均高于对照组(P<0.05);VD3干预后,两组IL-17A、IFN-γ、TNF和IL-2水平均低于干预前,且DN组IL-17A、IFN-γ和IL-2水平仍高于对照组,但TNF水平低于对照组(均P<0.05)。结论相关促炎细胞因子可能在DN发生中起促进作用,VD3对DN患者外周血单核细胞相关促炎因子具有抑制作用。 Objective To investigate the expressions of proinflammatory cytokines in the peripheral blood monocytes of patients with diabetic nephropathy before and after 1,25-dihydrovitamin D3 ( VD3 ) intervention. Methods Fifteen patients with type 2 diabetes mellitus complicated with diabetic nephropathy( DN group) and 15 healthy controls( control group) were enrolled. The levels of interleukin (IL)-17A,interferon( IFN)-3,, tumor necrosis factor(TNF) and IL-2 in culture supernatant containing peripheral blood monocytes were detected,respectively,in the two groups with and without VD3 intervention. Results Before VD3 intervention, there was no significant difference in the level of IL-17A between the two groups ( P 〉 O. 05 ), the levels of IFN-γ, TNF and IL-2 in the DN group were higher than those in the control group (P 〈 0. 05 ). After VD3 intervention, the levels of IL-17A, IFN-γ TNF and IL-2 in the two groups were all lower than those before intervention, and the levels of IL-17A, IFN-γ, and IL-2 in the DN group were higher than those in the control group, but the TNF level was lower than that in the control group ( all P 〈 0.05). Conclusion The relevant proinflammatory cytokines might play an acceleration role in the occurrence of DN, and VD3 has an inhibition effect on proinflammatory cytokines in the peripheral blood moncytes of DN patients.
