摘要
目的探讨不同浓度锌对脊髓损伤后抗氧化应激的内在联系。方法取70只SD大鼠,建立Allen's SCI模型,随机分成假手术组、模型组、低锌喂养组和高锌喂养组。HE染色检测各组大鼠脊髓损伤情况,BBB评分检测后肢运动功能的改善情况,TBA法测定丙二醛(MDA)含量和谷胱甘肽过氧化物酶(GSH-PX)活性,超氧化物歧化酶(SOD)试剂盒检测SOD活性。结果高锌喂养组GSH-PX和SOD表达量在每个时间点均明显高于其他3组,而SOD表达量在每个时间点均明显低于其他3组,组间差异有显著性意义(P<0.01)。术后2 w脊髓HE染色显示高锌喂养组脊髓损伤减轻,术后各时间点高锌喂养组大鼠BBB评分明显高于模型组和低锌喂养组。结论进食适量浓度锌可促进SCI大鼠GSH-PX和SOD高表达及MDA低表达,可明显改善大鼠抗氧化应激能力,促进脊髓损伤恢复。
Objective To investigate the effects of different concentration of zinc on the internal relationship of oxidative stressafter spinal cord injury. Methods Seventy SD rats were selected to build Allen's SCI model and were randomly divided into sham op-eration group, model group, low zinc feeding group and high zinc feeding group. HE staining was used to detect the rat spinal cord in-jury in every group, BBB score was used to detect the improvement of hind-limb motor function. TBA was used to measure MDA con-tent and activity of glutathione peroxidase (GSH-PX), Superoxide dismutase (SOD) kit was used to detect SOD activity. ResultsThe expression levels of GSH-PX and SOD in high-zinc-fed group were significantly higher than those in the other three groups at eachtime point, while the expression level of SOD was significantly lower than those of the other three groups at each time point. The differ-ences were significant (P〈O. 01 ). Postoperative 2 w spinal cord HE staining showed that spinal cord injury was reduced in high zincdiet group. BBB score of high zinc diet group was significantly higher than those of model group and low zinc diet group at each timepoint. Conclusion Supplement Zinc of proper concentration can improve the over expression of GSH-PX and SOD and the low ex-pression of MDA in SCI rats and also can significantly improve the ability of anti-oxidative stress and promote the recovery of spinalcord injury.
