中间普氏菌和脆弱类杆菌内毒素对人牙周膜细胞增殖和超微结构的影响

Effect of endotoxin of prevotella intermedia and bacteroides fragilis on proliferation and ultrastructure of periodontal ligament cell in vitro

目的 :观察中间普氏菌和脆弱类杆菌的内毒素对人牙周膜细胞增殖和超微结构的影响。方法 :体外培养人牙周膜细胞 ,MTT法测定中间普氏菌和脆弱类杆菌的内毒素对人牙周膜细胞增殖的影响 ,找出显效浓度 ,并应用透射电镜技术观察该浓度下两种内毒素对人牙周膜细胞超微结构的影响。结果 :两种内毒素在 10 μg/mL浓度时即能显著抑制人牙周膜细胞的增殖 ,该抑制作用随内毒素浓度增高而增大。 10 μg/mL浓度的内毒素使人牙周膜细胞的线粒体、内质网、高尔基氏体肿胀 ,溶酶体增多 ,核质浓集 ;甚至造成部分细胞死亡。结论 :中间普氏菌和脆弱类杆菌的内毒素可对人牙周膜细胞产生毒害作用 ,二者之间无明显差异。

AIM:To examine the effect of endotoxin of prevotella intermedia and bacteroides fragilis on proliferation and ultrastructure of periodontal ligament cells in vitro . METHODS: Human PDLC was cultured in vitro. MTT colormetic method was used to evaluate the effect of endotoxin on proliferation of PDLC. Transmission electron microscope was used to observe the ultrastructure changes of PDLC after treated with endotoxin. RESULTS:PDLC treated with 10 μg/mL endotoxin of prevotella intermedia or bacteroides fragilis showed a significant growth inhibition in dose-dependant manner. Marked ultrastarcture changes was found in the endotoxin treated cells, including mitochondria, endoplasmic reticulum and Golgi body swelling, lysosome increasing, nucleoplasm concentrating. Some cells even showed necrotic signs. CONCLUSION: Endotoxin of prevotella intermedia and bacteroides fragilis was harmful to PDLC and no significant difference was found between the cells treated with endotoxin of prevotella intermedia and bacteroides fragilis .
