去卵巢大鼠肿瘤坏死因子-α、降钙素、骨钙素与骨质疏松的关系 被引量:11

The relationship between serum levels of TNF-α,BGP,CT and osteoporosis of ovariectomized rats

摘要 目的通过观察卵巢切除后机体血清肿瘤坏死因子-α(TNF-α)、雌二醇(E2)、骨钙素(BGP)、降钙素(CT)的含量变化及其与骨密度之间的关系,探讨绝经后骨质疏松症的发病机理。方法以去卵巢大鼠为模型,定期检测模鼠骨密度和血清TNF-α、E2、BGP、CT的含量变化,并分析它们之间的相关性。结果术后12周模型组血清中的TNF-α、BGP浓度明显升高,而血清E2、CT浓度却下降,与假手术组相比均有统计学意义(P<0.05)。术后12周,模型组骨密度显著低于假手术组(P<0.005),而且随着术后时间的延长持续下降。相关分析显示:血清TNF-α、BGP与腰椎BMD负相关,而E2、CT与腰椎BMD正相关。结论当卵巢摘除E2水平下降时,骨代谢异常的病理机制是广泛而复杂的。其中TNF-α含量的增加,是骨吸收增强,发生骨质疏松不容忽视的原因之一。 Objective To investigate the mechanism of oste oporosis by observing the serum leve ls of TNF-á,E 2 ,BGP,CT and BMD of ovariectomized rats.Methods Osteoporosis model were e stablished by excising ovary of female rats.BM D were measured with dual energy X-ray absorptiometry and the levels of TNF-á,E 2 ,BGP,CT in serum were examined by ELI SA and RIA.Results The serum levels of TNF-á&BGP rose,but the E 2 &CT decreased in ovariectomized rats comparing with Sham group(P<0.05 ).The BMD decreased also(P<0.005).At the same time,the BMD were nega tively correlated with the serum levels of TNF-á,BGP,but positive correlated with t he E 2 ,CT.Conclusion The mechanism of bone metabolic diso rder was complicated a nd wide-ranging.The raising of TNF)áin serum was the one of reasons not to b e ignored in stimulating the bone resorption and bring about the osteoporosis.
