摘要
通过聚合酶链式反应(PCR)研究稀土离子对基因序列的体外诱变作用。以克隆在质粒上的乙酰化酶基因做模板进行PCR,在反应体系中加入不同浓度的稀土离子,并将PCR产物克隆测序。稀土离子对PCR有影响,当稀土离子的浓度达到1×10^(-3)mol/L时可以抑制PCR;将诱变后的乙酰化酶基因序列与原序列对比发现,稀土离子可以在体外通过PCR引起基因突变。突变是点突变即由T变为C和由G变为A的转换。
The aim of this research is to study the mutagenic effeet of rare earth ions on a gene sequencn. The plasmid containing the acetylation of enzyme gene was used as a template for PCR , different concentrations of rare earth ions were add- ed into the reaction system,the PCR products were cloned and sequenced. Rare earth ions can influence the PCR and inhibit the PCR when the concentration of rare earth ions reaches 1 × 10^-3mol/L. The obtained sequences were aligned with the origi- nal gene sequences,the results show that rare earth ions can cause gene mutations in vitro by PCR. The mutation is a point mutation - a single base change. The most mutations are transitions(e, g. GC→TA).
