自拟解酒护肝汤对酒精性肝炎模型大鼠的保护作用

Effects of Jiejiu Hugan Decoction on Alcoholic Hepatitis in Rats

目的探讨自拟解酒护肝汤对酒精性肝炎模型大鼠的肝脏保护作用。方法将SD大鼠随机分为6组,每组10只。将大鼠按体重用56%酒精给予灌胃,7 mL/kg,1天1次,持续4周。从造模后第5周起,阳性对照组灌胃给予西乐葆0.05 mg/kg,解酒护肝汤治疗组分别给予灌胃低、中、高剂量(5.0 mg/kg,15.0 mg/kg,30.0 mg/kg),持续30天。末次给药后,经腹主动脉取血,以及取肝脏组织,称重固定。随后试剂盒检测血清丙氨酸氨基转移酶(alanine aminotransferase,ALT)、谷氨酸氨基转移酶(aspartate transaminase,AST)和丙二醛(malondialdehyde,MDA),ELISA和免疫组化法分别检测血清和肝脏肿瘤坏死因子α(tumor necrosis factor,TNF-α),HE染色观察肝脏组织形态。结果与模型对照组比较,解酒护肝汤各剂量组的ALT、AST显著减少(P<0.05),解酒护肝汤中、高剂量组的MDA、TNF-α显著减少(P<0.05)。解酒护肝汤高剂量组的肝脏脏器系数显著下降(P<0.05)。解酒护肝汤各剂量组肝脏TNF-α表达均有一定程度减少。结论自拟解酒护肝汤对酒精性肝炎模型大鼠肝脏有一定的保护作用。

Objective To study the effects of Jiejiu Hugan Decoction on alcoholic hepatitis in rats. M ethods SD rats were randomly divided into 6 groups（n=10）. Model preparation was gastric gavage with 35% alcohol（ 15ml/kg） for 4 weeks,once a day. After models were built,positive control group was given celebrex 0.05 mg/kg,and protect liver soup group was given,Jiejiu Hugan decoction low,medium and high dose（ 5.0 mg/kg,15.0 mg/kg,30.0 mg/kg） for 30 days. After the last administration,blood was collected by abdominal aortic blood,and liver tissue was weighed and fixed. The serum ALT,AST and MDA were detected. TNF-α in serum and liver was detected by ELISA and immunohistochemical method. And the HE staining was used to observe liver. Results Compared with the model group,the level of ALT,AST,MDA and TNF-α of Jiejiu Hugan decoction was significantly reduce（P〈0.05）. The expressions of TNF-alpha and organ coefficients in liver were reduced in Jiejiu Hugan decoction groups and liver damage was slowed down. Conclusion Jiejiu Hugan decoction has a protect effect on alcoholic hepatitis in rats.
