肾动脉分支阻断技术对糖尿病小鼠缺血再灌注损伤及肾功能的保护作用 被引量:3

Protective effects of segmental renal artery clamping technique on ischemia-reperfusion injury in db/db diabetic mice

摘要 目的观察相对于传统的肾动脉阻断术(RAC),肾分支支脉阻断术(SRAC)对糖尿病小鼠(diabetic mice)缺血再灌注损伤及肾功能的保护性作用。方法糖尿病小鼠42只,随机分组为假手术组(Sham组)、肾动脉阻断组(RAC组)、肾分支动脉阻断组(SRAC组),每组14只。经过8周的饲养和观察后,将小鼠单侧肾蒂结扎,诱导出对侧孤立肾的模型。对3组小鼠按照不同的肾血管夹闭方式,诱导出不同的肾脏缺血再灌注损伤模型。在诱导肾脏缺血30min后,予以再灌注24h,分别取血样、尿样和肾皮质标本留作后续化验和组织学检查。结果术后24h,SRAC组小鼠的血肌酐、血尿素氮、尿微量白蛋白与尿肌酐比(UMAB/Ucr)显著低于RAC组(P<0.05)。结论 SRAC可有效地减少糖尿病小鼠缺血再灌注导致的早期肾脏损伤并加速肾功能的恢复。 ObjectiveTo investigate the protective effects of precise segmental renal artery clamping(SRAC) against renal I/R injury in diabetic mice compared with conventional renal artery clamping(RAC).MethodsA total of 42 diabetic mice were randomly divided into sham operation group(Sham group),RAC group and SRAC group,with 14 mice in each group.After 8 weeks,the unilateral renal pedicle was ligatured,and ischemia-reperfusion(I/R) injury to the contralateral kidney was induced.The 30min ischemia was followed by 24h reperfusion.Then the blood,urine and renal cortical samples were collected for subsequent laboratory tests and histological examination.ResultsBlood creatinine,blood urea nitrogen(BUN) and urine microalbumin/urine creatinine(UMAB/Ucr) ratio were significantly lower in the SRAC group than those in the RAC group 24h postoperatively.ConclusionSegmental renal artery clamping effectively reduces early renal injury induced by I/R and accelerates the recovery of renal function in diabetic mice.
