不同剂量螺内酯对高血压模型大鼠心肌保护作用分析 被引量:1

摘要 目的探讨不同剂量螺内酯对高血压模型大鼠的心肌保护作用。方法选择40只雄性SD大鼠,随机分为对照组、模型组、小剂量组和大剂量组各10只,模型组、小剂量组和大剂量组采用左旋精氨酸甲酯灌胃制作高血压模型,之后仍保持正常饮食,小剂量组给予20 mg/kg螺内酯灌胃,大剂量组给予40 mg/kg螺内酯灌胃,8周后处死,对各组大鼠的心肌病理学图像进行观察分析。结果心肌细胞横截面积、小动脉管壁/管腔面积比、心肌内小血管周围纤维化指数均为模型组>小剂量组>大剂量组>对照组,各组间对比差异均具有统计学意义(P<0.05),心肌组织胶原含量模型组、小剂量组和大剂量组均显著高于对照组(P<0.05),模型组显著高于小剂量组和大剂量组(P<0.05),但小剂量组和大剂量组之间对比无显著差异(P>0.05)。结论螺内酯能够有效保护高血压模型大鼠的心肌细胞,大剂量应用效果更好,但对心肌胶原含量的影响不大。 Objective To explore the effect of different doses of spironolactone on the myocardial protection in hypertension model rats. Methods A total of 40 male SD rats were randomly divided into control group, model group, low dose group and high dose group, with 10 rats in each group. In the model group, low dose group and high dose group, gavage of L-arginine methyl ester was used to prepare the hypertension model, and then the rats were maintained the normal diet. The low dose group was given 20 mg/kg of spironolactone garage, and the high dose group was given 40 mg/kg spironolactone gavage. The rats were executed after 8 weeks. The myocardial pathological images of each group of rats were observed and analyzed. Results The cross-sectional area of myocardial cells, the ratio of small arterial wall/lumen area, and the intramyocardial microvascular fibrosis index all ranked as follows: model group 〉 low dose group 〉 high dose group 〉 control group. The differences between groups were statistically significant(P〈0.05). The content of collagen in cardiac muscle tissue in the model group, low dose group and high dose group was all significantly higher than that in the control group (P〈0.05), and the model group was significantly higher than that in the low dose group and high dose group, but there was no significant difference between the low dose group and the high dose group(P〉0.05). Conclusion Spironolactone can effectively protect the cardiomyocytes of hypertension model rats. The high dose has a better application effect, but has little effect on the content of myocardial collagen.
