射干麻黄汤对急性哮喘小鼠模型IL-17A及IL-17F表达的影响

The Influence of Shegan Mahuang Decoction on the Expressions of IL-17A and IL-17F in Acute Asthma Mice

目的:探讨射干麻黄汤对急性哮喘小鼠模型肺组织及血清中IL-17A及IL-17F表达的影响,以近一步阐明其治疗哮喘的作用机制。方法:选取60只SD小鼠随机分为6组,分别为空白对照组(A组)、哮喘模型组(B组)、地塞米松对照组(F组)、射干麻黄汤高、中、低浓度组(C、D、E组)。显微镜镜下观察经HE染色的肺组织的病理性改变,哮喘小鼠血清检测IL-17A及IL-17F水平,肺组织中IL-17A mRNA及IL-17F mRNA的水平采用逆转录-聚合酶链反应法检测。结果:小鼠肺组织中B组IL-17A mRNA及IL-17F mRNA的表达量均明显高于A组(P<0.01)。与B组比较,C、D、E组BALF中IL-17A mRNA及IL-17F mRNA水平降低(P<0.05),且C组IL-17A mRNA及IL-17F mRNA表达量下降最为明显。C组与F组IL-17A mRNA及IL-17F mRNA表达量相接近。小鼠血清中IL-17A及IL-17F水平变化与小鼠肺组织IL-17A mRNA及IL-17F mRNA水平变化趋势基本相同。结论:射干麻黄汤可能是通过降低IL-17A及IL-17F细胞因子的分泌,来起到治疗哮喘的目的。

Objective： To investigate the influence of Shegan Mahuang Decoction on the expression of IL-17A and IL-17F in the lung tissue of acute asthma mice,and to further elaborate its mechanism of treating asthma. Methods： A total of 60 mice were randomly divided into six groups,namely the control group（ group A）,the asthma model group（ group B）,different concentration groups of Shegan Mahuang decotion（ group C,group D,and group E）,and Dexamethasone group（ group F）. The pathological changes of lung tissue were observed under microscope with HE staining method,levels of IL-17A and IL-17F were tested in serum,and the levels of IL-17A mRNA and IL-17F mRNA in lung tissue were detected by reverse transcription polymerase chain reaction. Results： The expressions of IL-17A mRNA and IL-17F mRNA in group B were significantly higher than those in group A（ P〈0. 01）. Compared to group B,they were both decreased in group C,group D,and group E（ P〈0. 05）,in which the changes in group C was most significant,closing to group F. The level changes of IL-17A and IL-17F had similar trend to the changes of IL-17A mRNA and IL-17F mRNA. Conlusion： Shegan Mahuang decoction may achieve the treating effect on asthma by reducing the production of IL-17A and IL-17F.
