针刺“百会”透“曲鬓”对急性脑出血大鼠血清中P选择素(CD62p)的表达影响的实验研究

Expression of CD62p in Acute Cerebral Hemorrhage Rats Intervened by Acupunture of Baihui-to-Qubin

目的:通过观察"百会"透"曲鬓"头针针刺法对急性脑出血大鼠脑组织中CD62p表达的影响,来揭示针刺头部腧穴对脑出血后炎症反应影响的相关机理方法:选取132只雄性Wistar大鼠随机分3组:空白对照组12只大鼠,脑出血模型组,针刺治疗组各60只大鼠。后两组又随机分为6 h,24 h,3 d,7 d四个亚组,每组大鼠数量均等。针刺组分别于相应的时间点进行针刺治疗,空白组,脑出血作为对照组不做处理。在完成治疗后,于相应的时间点对大鼠进行神经功能评分,然后处死大鼠提取血清,采用酶联免疫吸附法(ELISA)测定血浆CD62p表达水平。结果:经针刺治疗后,针刺组大鼠的神经功能缺损体征较同时间点的脑出血对照组明显减轻,差异具有统计学意义(P<0.01)。CD62p酶联免疫吸附法检测结果显示,脑出血组,针刺组CD62p水平较空白组明显增高,并有显著差异(P<0.01);针刺组脑组织中CD62p水平较相应时间段的脑出血组低,并有显著差异(P<0.01);脑出血组,针刺组在3d时脑组织中CD62p含量较其他时间段高,并有显著差异(P<0.01)。结论:"百会透曲鬓"针刺法可明显改善大鼠的神经功能学评分,有利于脑出血后肢体运动功能的恢复;"百会"透"曲鬓"针刺疗法,可能通过抑制CD62p在血小板膜上的表达,从而在一定程度上抑制脑出血后炎症反应,及脑水肿的形成。

Objective： To investigate the effectiveness of acupuncture to the inflammation after cerebral hemorrhage through observing the influence of the expression of CD62p after acute cerebral hemorrhage in rats by acupuncturing Baihui-to-Qubin. Methods： 132 Male Wistar rats were randomly divided into the blank group（ 12 rats）,the model group（ 60 rats）,and the acupuncture group（ 60 rats）,of which the last two groups were randomly and evenly divided into four sub-groups as different time point： 6 h,24 h,3 d,and 7 d. Acupuncture group was treated by acupuncture at different time,whereas the blank group and the model group were not intervened. After treatment,the scores of nerve function were assessed at different points of time,and serum levels of CD62p were detected in each group with ELISA method. Results： The neurological impairment was significantly alleviated after acupuncture treatment compared to the model group at the same time point（ P〈0. 01）. ELISA result： The expression of CD62p in the rats＇ brain which have cerebral hemorrhage was obviously higher than that of the blank group,and the difference was statistically significant（ P〈0. 01）; CD62p expression of the acupuncture group was obviously lower than that of the model group at the same time point（ P〈0. 01）; The expression of CD62p reached the peak on the 3rd day after operation,there was a significant difference compared to the other time point（ P〈0. 01）. Conclusion： Acupuncture of Baihui-to-Qubin can obviously raise the scores of nerve function,and have the benefit for improvement of extremity function after cerebral hemorrhage. Also,this treatment can suppress the expression of CD62p in the membranes of blood platelets,to restrain the formation of encephaledema and inhibit the inflammation reaction.
