姜黄素对小鼠寻常型银屑病模型TNF-α、IL-6和IL-8表达的影响

Impact study of curcumin on TNF-α,IL-6 and IL-8 level changes in serum of psoriatic mice model

目的观察姜黄素对小鼠寻常型银屑病模型的临床表现,皮损厚度以及皮损处TNF-α、IL-6和IL-8表达的影响。方法采用咪喹莫特制备小鼠银屑病皮损模型,观察并比较各组小鼠皮损模型处PASI积分情况、皮损模型处病理表现、表皮厚度以及银屑病动物模型皮损组织TNF-α、IL-6和IL-8的表达情况。结果三组小鼠在红斑、鳞屑、肥厚程度,PASI总分、表皮厚度、病理学、皮损组织TNF-α、IL-6和IL-8含量方面比较差异均有统计学意义,(P<0.05);其中,实验组与对照组对比,以上指标差异均具有统计学意义,(P<0.05)。实验组与空白组相比,除病理表现外,以上指标差异均无统计学意义,(P>0.05)。结论本研究发现姜黄素可有效改善寻常型银屑病小鼠模型的临床表现,皮损厚度以及TNF-α、IL-6和IL-8含量,推测姜黄素可能是通过调节TNF-α,减少炎症性细胞因子,抑制血管内皮细胞增生等作用,从而缓解皮肤过度角化而达到治疗银屑病的目的,本研究为临床应用姜黄素治疗银屑病提供一定依据。

Objective To observe the effect of curcumin on clinical manifestation, lesion thinkness and expression changes of TNF-α, IL-6 and IL-8 in skin lesions in mice model of psoriasis vulgaris. Methods Psoriatic mice model skin were prepared by imiquimod, then the PASI score, pathological manifes- tation, epidermis thinkness and expression levels of TNF-α, IL - 6 and IL - 8 of lesions were detected and compared between groups. Results There were statistically significant differences in erythema, scaling, hypertrophy degree, PASI score, epidermal thickness, pathology, TNF- alpha, IL-6 and IL-8 contents in three groups of mice （ P〈0. 05 ） ; Among them, the above index were statistically significant （ P〈 0. 05 ）. But, between experimental group and blank group, exept symptoms, no statistically significant difference were observed in the above indicators, （ P〉 0.05 ）. Conclusion This study found that curcumin can effectively improve the clinical manifestations of psoriasis mice model, thinkness of lesion, and content of TNF- alpha, IL-6 and IL-8, which may attribute to the regulation of TNF- alpha, reduction of inflammatory cytokines and inhibition the proliferation of vascular endothelial cells by curcumin, leading to alleviate the skin hyperkeratosis, then achieve the purpose of the treatment of psoriasis. This study provides a basis for the clinical application of curcumin in the treatment of psoriasis.
