润燥灵对干燥综合征模型小鼠免疫调节及抗炎作用研究 被引量:3

On the Immunoregulation and Anti-inflammation of Runzao Ling(润燥灵)on Model Mice with Sj?gren's Syndrome

摘要 目的:探讨中药润燥灵方对干燥综合征模型小鼠免疫器官的影响及其抗炎作用。方法:将90只C57bl/6j小鼠随机分为空白对照组,模型对照组,醋酸泼尼松组和润燥灵高、中、低剂量组,每组15只。除空白对照组外,其余各组采用免疫诱导法建立干燥综合征小鼠模型,造模第5周开始灌胃给药。空白对照组、模型对照组均给予蒸馏水0.2 mL·d^(-1)灌胃,醋酸泼尼松组给予醋酸泼尼松混悬液2 mg·kg^(-1)·d^(-1)灌胃,润燥灵高、中、低剂量组分别给予润燥灵50,25,12.5 g·kg^(-1)·d^(-1)灌胃。用药4周后眼球取血,乌拉坦麻醉后断颈处死小鼠,分离颌下腺、胸腺和脾脏,计算小鼠颌下腺、胸腺和脾脏指数,HE染色观察病理变化,ELISA法检测小鼠血清白细胞介素-17(IL-17)的分泌情况。结果:颌下腺指数研究结果显示,润燥灵高剂量组小鼠颌下腺指数较模型对照组明显增加(P﹤0.05),与醋酸泼尼松组相当(P﹥0.05)。胸腺和脾脏指数研究显示,与模型对照组比较,润燥灵高剂量组和醋酸泼尼松组均能明显降低干燥综合征小鼠胸腺和脾脏指数(P﹤0.05或P﹤0.01),而中、低剂量组影响不明显(P﹥0.05)。病理结果研究显示,与模型对照组比较,润燥灵高剂量组能够明显改善干燥综合征小鼠颌下腺、胸腺和脏脾的病理变化,而中、低剂量组不明显。ELISA检测结果显示,模型对照组小鼠IL-17分泌量增加(P﹤0.01);与模型对照组比较,润燥灵高、中剂量组均能降低IL-17的分泌量(P﹤0.05),低剂量组降低不明显(P﹥0.05)。结论:润燥灵具有降低干燥综合征模型小鼠颌下腺指数,下调IL-17分泌的作用。 Objective:To investigate the effects of Runzao Ling (润燥灵 ) on immune organs in mice with sj6gren's syndrome and its anti-inflammatory effects.Methods:Ninety C57bl/6j mice were randomly divided into a blank control group,a model control group,a prednisone acetate group and Runzao Ling groups (with high,medium and low doses ) ,15 rats in each group.Except for the blank control group,the mice in other groups were induced by immune induction to establish the models with sjogren's syndrome,and the models were given intragastric administration from the fifth week.The rice in the blank control group and the model control group were given intragastric administration of distilled water ( 0.2 mL d-1 ) .The mice in the prednisone group were given intragastric administration of prednisone suspension ( 2 mg kg-1 d-1 ) . The mice in the Runzao Ling groups (with high,medium and low doses )were given intragastric administration of Runzao Ling ( 50,25,12.5 g kg-1 d-1 respectively ) .After 4 weeks of treatment,the eye blood was taken and the urethane anesthetized mice were killed.The submandibular gland,thymus and spleen of the killed mice were separated to calculate their index.Their pathological changes were observed by HE staining and the ELISA method was used to detect the secretion of IL-17 in the serum.Results:The submandibular gland index in the high-dose Runzao Ling group were significantly higher than those in the model control group ( P 〈 0.05 ) ,and were almost equivalent to those of the prednisone group ( P 〉 0.05 ) .Compared with the model group,the high-dose Runzaoling group and the prednisone group could significantly reduce the index of thymus and spleen in mice with sj6gren's syndrome ( P 〈 0.05 or P 〈 0.01 ) ,while there was no obvious effects in the medium and low dose groups ( P 〉 0.05 ) .The pathological results showed that compared with the model control group,the high-dose Runzao Ling group could obviously improve the pathological changes of the submandibular gland,thymus and spleen in the mice with sj6gren's syndrome,but those in the middle and low dose groups are not obvious.ELISA test results showed that IL-17 secretion of mice in the model group increased ( P 〈 0.01 ) .Compared with the model group,medium and high dose groups of Runzao Ling could reduce the secretion of IL-17 (P 〈 0.05 ) ,while that in the low-dose group did not significantly decrease (P 〉 0.05 ) . Conelusion:Runzao Ling can reduce the submandibular gland index and down regulate the secretion of IL-17 in model mice with sjogren's syndrome.
