Determination of Fructose, Glucose and Sucrose by Pressurized Capillary Electrochromatography Coupled to Evaporative Light Scattering Detector

摘要 本实验建立了食品中果糖,葡萄糖和蔗糖的亲水作用色谱的pCEC-μELSD分析方法,并进行了方法学验证和准确度测试。该方法在pCEC-μELSD联用平台上使用TSK Amide-80毛细管色谱柱(150μm I.D.×150 mm,5μm),在80%乙腈水(含30 mmol三乙胺),分离电压+5 KV条件下,实现了果糖,葡萄糖和蔗糖的快速分离。实验结果表明,方法线性范围为0.01~1.00μg;检出限4 ng,回收率在88.1%~103.9%之间;RSD在0.9%~3.3%之间。本方法为食品中糖类物质提供了一种简单快速的检测新方法。 A method base on pCEC-μELSD was developed for the determination of a mixture of fructose, glucose and sucrose in food, TSK Amide-80 capillary column (150 mm ×150 μm I.D., 5 μm) was selected. The analytes can be separated within 12 min under + 5 kV of applied voltage. The mobile phase was chosen as 80 % acetonitrile in DI water. The linear range of the method was from 0.01 to 1.0/ag for each analytes. The detection limit was 4 ng. Recoveries were from 88.1% to 103.9 % with RSD of 0.9 %-3.3 % (n=6), indicating that adaptability and stability of pCEC with μELSD are comparable with commercial HPLC-ELSD. This method provides a simple and rapid method for the determination of carbohydrates in foods.
