摘要
目的用紫外分光光度法测定铁包金的总黄酮含量。方法用80%的乙醇提取铁包金的总黄酮。以芦丁为标准品,测定其270 nm处吸光度。结果芦丁检测浓度在0.00424~0.02544 mg/m L范围内与吸光度呈现良好的线性关系,R=0.9990,平均加样回收率为99.47%,RSD=0.19%(n=5);用乙醇回流提取法提取的总黄酮含量可达到0.81%。结论:本方法操作简便,线性关系良好,准确度、重复性和稳定性较好,适用于瑶药铁包金黄酮类物质总含量的快速测定。
Objective:To determine the total flavonoids in the roots ofBerchemia lineata by UV-spectrophotometry. Method: The total flavonoids were extracted by 80 % ethanol from the roots of Berchemia lineata. Rutin was used as standard and the absorbances were detected at 270 nm. Results: Standard curve was established in the range of 0.00424-0.02544 mg/mL for total flavonoids, R=0.9990, and the average recovery was 99.82 %. The RSD of the method was 0.16 %, and the content of total flavonoids was 0.805 %. Conclusion: This method is simple, accurate and reproducible with good linearity. It can be used for rapid determination of total flavonoids of Berchemia lineata.
