摘要
目的通过高脂喂养与颈动脉内膜干燥建立餐后低血压的家兔模型。方法 17只新西兰兔随机分四组,对照组普通饲料喂养,高脂喂养组予高脂饲料喂养,高脂喂养+单侧手术组予高脂喂养并进行右侧颈动脉气体干燥,高脂喂养组+双侧手术组予高脂喂养并进行双侧气体干燥。8周后留置胃管,行股动脉插管,监测注食前后各组家兔血压变化。结果光镜下染色观察:对照组颈部血管无明显变化,高脂喂养组内膜轻度增厚,可见血管平滑肌细胞的增生。高脂喂养+单侧手术组的右侧(手术侧)颈动脉及高脂喂养+双侧手术组的双侧颈动脉管壁明显增厚,管腔狭窄。高脂喂养+单侧手术组注食后平均动脉压(Mean arterial pressure,MAP)较注食前下降(P<0.05);高脂喂养+双侧手术组注食后收缩压(Systolic blood pressure,SBP)、舒张压(Diastolic blood pressure,DBP)、MAP均较注食前下降,差异有统计学意义(P<0.05)。结论利用高脂喂养与颈动脉内膜干燥结合可建立餐后低血压家兔模型,此方法简单可行。
Objective To establish a rabbit model of atherosclerosis postprandial hypotension by using cholesteroldiet and carotid intimal dryness. Methods Seventeen New Zealand rabbits were assigned into four groups randomly: normal group fed with normal food, lipo-fed group fed with high lipo food,lipo-fed and unilateral operation group fed with high lipo food and received an operation with right side carotid artery air-drying,lipo-fed and bilateral operation group fed with high lipo food and received an operation with bilat-eral carotid artery air-drying. After 8 weeks,the indwelling gastric tube and femoral artery catheterization were used to monitor the blood pressure changes of rabbits before and after the feeding. Results Trypan-blue staining was observed by light microscopy, there were no obvious changes in the blood vessels of control group; the intima was slightly thickened with proliferation of vascular smooth muscle cells in the high-lipo diet group. In the group of lipo-fed and bilateral operation and the group of lipo-fed and unilateral opera-tion, the arterial wall was thickened and luminal stenosis was observed. In the group of lipo-fed and unilateral operation, the mean ar-terial pressure ( MAP ) was declined after the feeding ( P 〈 0 .0 5 ). In the group of lipo-fed and bilateral operation,the SBP, DBP, MBP were all decreased statistically after the feeding ( P 〈 0 . 0 5 ) . Conclusions A rabbit model of atherosclerosis postprandial hypotension can be established by combination of high fat feeding and carotid intima drying, which is simple and feasible . which advan-taged the experimental study of postprandial hypotension.
