解酒饮对急性胃黏膜损伤小鼠的保护作用

Protective Effect of Jiejiu Oral Liquid on Acute Gastric Mucosal Injury in Mice

目的探讨解酒饮对急性胃黏膜损伤小鼠的保护作用。方法昆明种小鼠60只,随机分为正常对照组、模型对照组、硫糖铝组(600 mg·kg-1)和解酒饮小、中、大剂量(6.25,12.5,25.0 g·kg-1)组,每组10只。除正常对照组外,其余各组给予95%乙醇10 m L·kg-1灌胃造模,测定各组小鼠胃黏膜损伤指数和抑制率,测定胃黏膜中一氧化氮(NO)、内皮素-1(ET-1)、肿瘤坏死因子α(TNF-α)、白细胞介素-6(IL-6)和前列腺素E2(PGE2)的含量。同时做组织病理学检查。结果模型对照组小鼠胃黏膜出现明显出血、糜烂和点片状溃疡。与模型对照组比较,解酒饮组急性胃黏膜损伤指数明显降低,中、大剂量组胃黏膜ET-1、TNF-α和IL-6含量降低(P<0.05或P<0.01)。解酒饮大剂量组胃黏膜NO水平升高(P<0.05)。解酒饮对胃黏膜PGE2含量无明显影响(P>0.05)。病理组织检查发现解酒饮可减少胃黏膜的脱落和炎症细胞浸润。结论解酒饮具有保护急性胃黏膜损伤小鼠的作用。

Objective To investigate the protective effect of Jiejiu oral liquid on acute alcohol-induced gastric mucosal lesions in mice. Methods Sixty Knming ' s mice were randomly divided into normal control group,model control group,sucralfate group( 600 mg·kg-1),the dose of 25.0 g·kg-1group,the dose of 12.5 g·kg-1group and the dose of 6.25 g·kg-1group of the Jiejiu oral liquid( n = 10 each).Except for the normal control group,other groups were intragastrically administered with 95% ethanol to create the alcohol-induced acute gastritis model. The effect of pathological histology,injury of gastric mucosa,the mucosal NO,ET-1,TNF-α,IL-6 and PGE2 content of acute alcohol-induced gastric mucosal lesions in mouse were observed. Results The gastric mucosa in model control group appeared obvious bleeding,erosion,and flake ulcer. Compared with the model control group,the acute damage index of the Jiejiu oral liquid therapy groups were obviously reduced( P<0.05 or P<0.01). The content of ET-1,TNF-α and IL-6 in the 25.0 g·kg-1and the 12.5 g·kg-1dose group of the Jiejiu oral liquid significantly decreased compared with the model control group( P < 0. 05 or P < 0. 01). The content of NO in the 25. 0 g · kg-1group of the Jiejiu oral liquid increased( P<0.05). Jiejiu oral liquid had no significant effect on the content of PGE2( P>0.05).Histopathologic examination revealed Jiejiu oral liquid can reduce the shedding of gastric mucosa and inflammatory cells infiltration. Conclusion The Chinese herbal mixture Jiejiu oral liquid has a protective effect on acute alcohol-induced gastric mucosal lesions in mice.
