G蛋白偶联受体5在脑胶质瘤病理组织中的表达及临床意义

摘要 目的 探讨G蛋白偶联受体5(LGR5)分子在脑胶质瘤病理组织的表达及其意义.方法 选择由不同病理分级脑胶质瘤组织以及正常脑组织构建的组织芯片,以抗人LGR5抗体和辣根过氧化物酶标记的抗大鼠抗体进行免疫组织化学染色,采用Aperio病理扫描系统结合两位病理科医师进行读片和结果分析.结果 LGR5分子在正常脑组织呈弱表达;脑胶质瘤组织中LGR5的表达明显高于正常脑组织;LGR5在病理Ⅰ级癌瘤组织和正常脑组织表达无明显差异;LGR5在病理Ⅱ、Ⅲ和Ⅳ级癌瘤组织表达明显增高,并明显高于正常脑组织和病理Ⅰ级脑胶质瘤组织.结论 LGR5高表达于脑胶质瘤组织,可能是脑胶质瘤病理诊断的潜在标记分子. Objective To explore the expression of LGR5 on glioma and its clinical significance. Methods Tissue chip that consisted of the tumor tissue from 57 glioma patients and normal brain tissue from 3 individuals was stained with anti-human LGR5 antibody and HRP labeled anti-rat antibody. The tissue chip was evaluated by Aperio pathological scan system and two experienced pathologist. Results LGR5 weakly expressed on normal brain tissues, and expressed highly on the glioma tissues. There is no significant difference between LGR5 expression on WHO grade Ⅰ glioma tissue and that on normal brain tissue. LGR5 expressed higher on WHO grade Ⅱ, Ⅲ and Ⅳ glioma tissue than that on WHO grade Ⅰ glioma tissue or normal brain tissue. Conclusion LGR5 expressed on glioma tissues in higher level and it is the promising indicator for glioma diagnosis.
