摘要
目的:建立同时测定补骨脂药材中补骨脂二氢黄酮和补骨脂酚含量的方法。方法:采用高效液相色谱法。色谱柱为Shim-Pack VP-ODS,流动相为乙腈-水(梯度洗脱),流速为1.0 mL/min,检测波长为246 nm,柱温为30℃,进样量为10μL。结果:补骨脂二氢黄酮、补骨脂酚检测进样量线性范围分别为48.9~489 ng(r=0.999 8)、784~7 840 ng(r=0.999 9);精密度、稳定性、重复性试验的RSD<2.0%;加样回收率分别为95.30%~99.63%(RSD=1.45%,n=9)、96.89%~100.82%(RSD=1.36%,n=9)。结论:该方法操作简便,精密度、稳定性、重复性好,可用于补骨脂药材中补骨脂二氢黄酮和补骨脂酚含量的同时测定;不同辐照剂量下药材样品中待测成分的含量均无明显变化。
OBJECTIVE: To establish a method for content determination of bavachin and bakuchiol in Psoralea corylifolia. METHODS: HPLC method was adopted. The determination was performed on Shim-Pack VP-ODS column with mobile phase con- sisted of acetonitrile-water (gradient elution) at the flow rate of 1.0 mL/min. The detection wavelength was set at 246 nm, the col- umn temperature was 30 ℃ , and the sample size was 10 μL. RESULTS: The linear ranges of bavachin and bakuchiol were 48.9-489 ng(r=0.999 8), 784-7 840 ng(r=0.999 9). RSDs of precision, stability and reproducibility tests were all lower than 2.0%. Average recoveries ranged 95.30%-99.63% (RSD=l.45%, n----9), 96.89%-100.82% (RSD=1.36%, n=9). CONCLU- SIONS: The method is simple, accurate, stable and reproducible, and can be used for the simultaneous determination of bavachin and bakuchiol in P. corylifolia.
