清脑止痛胶囊对利血平致偏头痛模型大鼠的影响 被引量:6

摘要 目的研究清脑止痛胶囊(QNZTJN)对利血平致偏头痛模型大鼠血浆和中脑组织相关神经递质及炎性因子的影响。方法 90只健康雄性Wistar大鼠,随机均分为6组:空白组,模型组,阳性药正天丸组(ZTW,0.006g/kg),清脑止痛胶囊剂量低(QNZTJN-L,0.35g/kg)、中(QNZTJN-M,0.7g/kg)、高(QNZTJN-H,1.4g/kg)组。皮下注射利血平(0.7mg/kg)14d建立偏头痛模型,造模第4d开始灌胃给药11d,观察末次给药后3h内大鼠行为学反应;免疫组织化学法及Western blot法,检测大鼠中脑组织核因子kappa B(NF-κB)和环氧合酶2(COX-2)表达水平;酶联免疫法,检测大鼠血浆和中脑组织5-羟色胺(5-HT)、前列环素E_2(PGE_2)、肿瘤坏死因子α(TNF-α)、白介素1β(IL-1β)含量水平。结果模型组与空白组相比,大鼠挠头次数显著增加(P<0.01),NF-κB、COX-2在中脑组织表达显著增加(P<0.01),5-HT在血浆和中脑组织含量显著降低(P<0.01),PGE_2、TNF-α、IL-1β在血浆和中脑组织含量显著升高(P<0.05,P<0.01)。QNZTJN-M、QNZTJN-H组和ZTW组与模型组相比,大鼠挠头次数显著减少(P<0.05,P<0.01),NF-κB、COX-2在中脑组织表达显著降低(P<0.05,P<0.01),5-HT在血浆和中脑组织含量显著升高(P<0.05,P<0.01),PGE_2、TNF-α、IL-1β在血浆和中脑组织含量显著降低(P<0.05,P<0.01)。结论清脑止痛胶囊可缓解利血平致大鼠偏头痛症状,其作用机制可能与调节神经源性炎症和单胺类神经递质释放有关。 Objective To detection the effects of QNZTJN on migraine rat model induced by reserpine.MethodsTotally 90 Wistar rats(males)were randomly divided into normal group,model group,ZTW positive group(0.006 g/kg),QNZTJN high dose group(1.4 g/kg),QNZTJN medium dose group(0.75 g/kg),QNZTJN low dose group(0.35 g/kg).The migraine model rats were induced by subcutaneous injection 0.7 mg/kg reserpine to observe the forelimb scratching frequency and the effects of different concentrations of QNZTJN on the level of NF-κB,COX-2,5-HT,PGE_2,TNF-αand IL-1β in the midbrain tissue and plasma.Results Compared with the normal group,the forelimb scratching frequency were increased significantly in model group(P〈0.01).The contents of COX-2 and NF-κB in midbrain tissue were increased significantly(P〈0.01).The contents of PGE_2,TNF-αand IL-1β were increased,on the contrary the contents of 5-HT was decreased in both midbrain tissue and plasma(P〈0.05,P〈0.01).Compared with model group,QNZTJN medium and high dose significantly decreased the forelimb scratching frequency of rats(P〈0.05,P〈0.01).The contents of COX-2 and NF-κB in midbrain tissue were decreased significantly(P〈0.05,P〈0.01).The contents of PGE_2,TNF-αand IL-1β were decreased,while the contents of 5-HT was increased in both midbrain tissue and plasma(P〈0.05,P〈0.01).Conclusion The QNZTJN can relieve migraine attacks induced by reserpine,which may be related to regulating the release of monoamine neurotransmitters and neurogenic inflammation.
