响应面法优化灵芝总三萜酶辅助提取工艺及其抗氧化活性研究 被引量:25

Response surface optimization of enzyme-assisted extraction triterpene from Juncao Ganoderma lucidum and its antioxidant activity

摘要 利用响应面法对灵芝总三萜的提取工艺进行优化,同时对其抗氧化活性进行研究。在单因素酶解时间、酶解p H、酶解温度和酶用量的基础上,选取纤维素酶、果胶酶和中性蛋白酶的酶用量比例为自变量,灵芝总三萜得率为响应值,进行响应面试验设计,得到最优提取条件为:酶解时间为80 min、提取p H为5、酶解温度50℃、复合酶用量比例为纤维素酶:果胶酶:中性蛋白酶为2.11:0.86:2.09,在该条件下预测值为1.02%、实际值为0.99%、相对误差为2.9%。抗氧化试验中灵芝提取液显示出对DPPH自由基和羟基自由基有一定的清除能力,同时具有较好的还原能力。 The triterpene extraction from Ganoderma lucidum was optimized by response surface, and the antioxidant activities were investigated in the paper. Based on the single factors of extraction time, extraction pH, extraction temperature and the concentration of enzyme, the concentration ratio of three enzyme, including cellulase, neutral protease and pectinase, was chose as independent variable, and the yield of trterpene was the response value. The results were as followings: extraction time 80 min, extraction pH 5 extraction temperature 50 ℃, and the concentration ratio of three enzyme was cellulose:pectinase:neutral protease of 2.11:0.86:2.09. Under the optimum condition, the predictive yield was 1.02%, and actual yield was 0.99%, the relative error was 2.9%. The water extraction from Ganoderma lucidum showed the scavenging activity on the DPPH and hydroxyl radicals and possessed a great reducing power.
