摘要
建立了一种食品中去甲肾上腺素和多巴胺的液相色谱定量分析及液相色谱-质谱/质谱确证方法。采用XBridge?C18(4.6 mm×250 mm,5μm)色谱柱,柱温30℃,流动相为v(磷酸盐缓冲液):v(乙腈)=98:2,流速0.4 m L/min,荧光检测器检测,激发波长为286 nm,发射波长为318 nm。结果表明,去甲肾上腺素在(0.02~50)μg/m L,多巴胺在(0.05~50)μg/m L内,其峰面积与质量浓度呈良好的线性关系(0.9999和0.9996),并采用超高效液相-质谱/质谱在复杂基质情况下辅助定性。该方法的检出限为去甲肾上腺素1.0 ng/m L,多巴胺2.0 ng/m L。该方法快速简便、灵敏度高、选择性好,测定结果令人满意。可将该方法应用到食品中去甲肾上腺素和多巴胺的测定中。
A HPLC method for the determination of both noradrenaline and dopamine was established, and the UPLC/MS/MS was developed to help with the qualitative analysis in the complex matrix. The HPLC was carried out on a 30 ℃ thermostatic XBridge C18 column (4.6 mm×250 mm, 5 μm). The mobile phase was consisted of v(phosphate buffer):v(acetonitrile)=98:2, at a flow rate of 0.4 mL/min. The detection was conducted by fluorescence detector with excitation wavelength of 286 nm and emission wavelength of 318 nm. Results showed that as the concentration of noradrenaline was in the range of (0.02N50)pg/ mL, and the concentration of dopamine was in the range of (0.05N50)lJg/mL, the realationship between concentration and the corresponding peak area showed good linearity (R=0.9999 and 0.9996). The UPLC/ MS/MS was used to qualitatively analyze in the complex matrix. The detection limit reached 1.0 ng/mL, 2.0 ng/mL for noradrenaline and dopamine. The method is easy, quick, sensitive and suitable for determination of both noradrenaline and dopamine in foods.
